Gastrointestinal stromal tumours (GISTs) are a precise oncogenetic entity that has started the new era of targeted therapies in solid tumours. GISTs are now a model to understand the role of oncogenic kinase mutation in tumourigenesis, duplication, cell regulation, apoptosis and drug resistance. Ninety-five percent of GISTs have some activacting mutation of c-KIT or PDGFRA tyrosine kinase. The studies of the last two years have found concrete correlations between mutations and anatomical locations of GISTs, prognosis, response to therapy and resistance to therapy. Genotyping has increased of importance in the last years as a new field for translational researches. The important advances made in molecular studies are now a practical tool in diagnosis and therapy of GISTs.

Advanced Targeted Therapy for Colorectal Cancer with Lipid Nanoparticles.

Curr Med Chem

January 2025

Department of Pharmaceutics, Shree S. K. Patel College of Pharmaceutical Education and Research, Ganpat University, Gujarat, Kherva, 384012, India.

Targeted therapy for colorectal cancer (CRC) appears to have great potential with lipid nanoparticles (LNPs). The advances in LNP-based techniques, such as liposomes, exosomes, micelles, solid lipid nanoparticles (SLNs), nano-cubosomes, and plant- derived LNPs (PDLNPs), are explored in detail in this thorough review. Every platform provides distinct advantages: liposomes enable precise drug release and improved delivery; exosomes function as organic nanocarriers for focused treatment; SLNs offer greater stability; micelles enhance drug solubility and resistance; nano-cubosomes tackle low bioavailability; and PDLNPs offer biocompatible substitutes.

Background: Neurofibromatosis type 1 (NF-1), a rare autosomal dominant disorder, arises from gene mutations affecting neurofibromin, a Ras GTPase regulator. These mutations activate Ras proteins, triggering clinical symptoms such as skin spots, epilepsy, pain, and tumors. Although gastrointestinal stromal tumors are well-known in NF-1, diffuse intestinal ganglioneuromatosis remains an extremely rare complication.

Investigation of unfavorable prognostic factors for survival in Chinese patients with gastric gastrointestinal stromal tumors.

Transl Cancer Res

December 2024

Key Laboratory of Tropical Translational Medicine of Ministry of Education, the First Affiliated Hospital, Hainan Medical University, Haikou, China.

Background: Gastrointestinal stromal tumor (GIST) was very rare in the gastrointestinal (GI) tract. Most GISTs were asymptomatic at early stage. Therefore, it was of great significance to explore the prognostic factors of patients with GIST.

Laparoscopic wedge resection of a descending duodenal gastrointestinal stromal tumor under endoscopic nasobiliary drainage guidance: A case report.

Int J Surg Case Rep

January 2025

Department of Surgery, Inje University Haeundae Paik Hospital, 875 Haeundae-ro, Haeundae-gu, Busan 48108, Republic of Korea. Electronic address:

Introduction: Gastrointestinal stromal tumors (GIST), which occur anywhere in the gastrointestinal (GI) tract, typically occur in the stomach and small intestine but rarely in the duodenum. We present a case report wherein a descending duodenal GIST was treated with a limited, minimally invasive surgery after endoscopic nasobiliary drainage (ENBD) insertion.

Presentation Of Case: A 67-year-old woman visited our hospital with an incidentally discovered duodenal tumor.
