Objective: To study systematically the factors which affect separation and purification of the total alkaloids and mesaconitine with X -5 macroporous resin.

Method: With the content of the total alkaloids and mesaconitine as parameters, the optimum condition of absorption and elution were studied in the process of the purification with X -5 macroporous resin.

Result: The X - 5 macroporous resin yielded the best separating efficiency when the concentration of the extracted solution was 1 g raw material per 1 mL, pH 12.0, the absorptive time of 6 hour and the volume of 95% ethanol (7BV pH 8) as the eluant; X -5 macroporous resins was used five times in a reproducible way. The rate of extraction and content of the total alkaloid were 80% and 30% respectively after purification with X - 5 macroprous resin.

Conclusion: The method can increase the purity of mesaconitine and total alkaloids.

Enantioselective Total Syntheses of Vallesamidine and Schizozygane Alkaloids.

J Am Chem Soc

January 2025

Department of Chemistry and Biochemistry, The University of Texas at Dallas, Richardson, Texas 75080, United States.

A general streamlined strategy for the enantioselective total syntheses of the schizozygane family of natural products and related alkaloid vallesamidine is described. Specifically, a catalytic enantioconvergent cross-coupling sets the quaternary stereogenic center in a pluripotent intermediate adorned with an olefin and three orthogonal carboxylate groups, upon which the modularity of the synthesis relies. A late-stage catalytic oxidative lactamization of an alkyne is instrumental in the first-generation synthesis of the schizozygane alkaloids.

The absolute and relative configurations of bioactive chiral molecules are typically relevant to their biological properties. It is thus highly important and desirable to construct all possible stereoisomers of a lead candidate or a given bioactive natural compound. Synergistic dual catalysis has been recognized as a reliable synthetic strategy for a variety of predictable stereodivergent transformations.

Selenium is a beneficial element in agriculture, particularly for its potential to improve plant growth and stress tolerance at suitable concentrations. In this study, Phaseolus vulgaris was foliar-sprayed with selenium selenate (Se) or selenium nanoparticles (SeNP) at different concentrations during the vegetative stage; afterward, the seed yield was analyzed for metabolomics using H, J-resolved and HSQC NMR data, and NMR databases. A total of 47 metabolites were identified with sugars being the major chemical class.
