The inhibitory activity of ovomucoid from duck egg white, immobilized on chitosan with the use of glutaraldehyde or carbodiimide as cross-linking agents, was studied. Glutaraldehyde proved to be a more preferable cross-linking agent than carbodiimide. When chitosan is used as a protein carrier, the possibility of shifting the pH optimum of these compounds should be taken into account.

The effects of hydrolysis by free and immobilized forms of Neutrase (FN, IN, respectively) and Thermolysin (FT, IT, respectively) and in vitro digestion on the degree of hydrolysis (DH) of egg white proteins, molecular weight distribution of peptides, immune-reactivity and IgE epitopes of egg white proteins were investigated. With FT and IT in the intestinal digests, the proteolysis followed by in vitro digestion produced peptides smaller than 10 kDa. Gal d 1 (ovomucoid) is the dominant allergen in the chicken egg white. Hypoallergenic variants of this allergen can be used in immunotherapy as an egg allergy treatment approach. We hypothesised that disruption of two of the nine cysteine-cysteine bridges by site-directed mutagenesis will allow the production of a hypoallergenic variant of the protein; Methods: Two cysteine residues at C192 and C210 in domain III of the protein were mutated to alanine using site-directed mutagenesis, to disrupt two separate cysteine-cysteine bridges.

A method has been developed for producing a biospecific hydrogel hemosorbent by the radical copolymerization of an unsaturated derivative of ovomucoid from duck egg white with acrylamide and N,N'-methylene bisacrylamide in an aqueous solution in the presence of mercaptoacetic acid serving as a chain transfer agent. The use of a chain transfer agent has been shown to result in changes in the structure of the hydrogel formed, namely, an increase in the degree of swelling in aqueous solutions and a decrease in the number of large pores. This creates favorable conditions for the functioning of immobilized ovomucoid and allows for an increase in the serine proteinase absorption capacity of the hemosorbent.

An optical immunochip biosensor has been developed as a rapid method for allergen detection in complex food matrixes, and its application evaluated for the detection of the egg white allergens, ovalbumin and ovomucoid. The optical near-field phenomenon underlying the basic principle of the sensor design is called resonance-enhanced absorption (REA), which utilizes gold nanoparticles (Au NPs) as signal transducers in a highly sensitive interferometric setup. Using this approach, a novel, simple, and rapid colorimetric solid-phase immunoassay on a planar chip substrate was realized in direct and sandwich assay formats, with a detection system that does not require any instrumentation for readout.
