Background: Hyperhomocysteinaemia (HHC) is thought to be a risk factor for cardiovascular disease including heart failure. While numerous studies have analyzed the role of homocysteine (Hcy) in the vasculature, only a few studies investigated the role of Hcy in the heart. Therefore we have analyzed the effects of Hcy on isolated cardiomyocytes.
Methods: H9c2 cells (rat cardiomyoblast cells) and adult rat cardiomyocytes were incubated with Hcy and were analyzed for cell viability. Furthermore, we determined the effects of Hcy on intracellular mediators related to cell viability in cardiomyocytes, namely NOX2, reactive oxygen species (ROS), mitochondrial membrane potential (DeltaPsi (m)) and ATP concentrations.
Results: We found that incubation of H9c2 cells with 0.1 mM D,L-Hcy (= 60 microM L-Hcy) resulted in an increase of DeltaPsi (m) as well as ATP concentrations. 1.1 mM D,L-Hcy (= 460 microM L-Hcy) induced reversible flip-flop of the plasma membrane phospholipids, but not apoptosis. Incubation with 2.73 mM D,L-Hcy (= 1.18 mM L-Hcy) induced apoptosis and necrosis. This loss of cell viability was accompanied by a thread-to-grain transition of the mitochondrial reticulum, ATP depletion and nuclear NOX2 expression coinciding with ROS production as evident from the presence of nitrotyrosin residues. Notably, only at this concentration we found a significant increase in S-adenosylhomocysteine which is considered the primary culprit in HHC.
Conclusion: We found concentration-dependent effects of Hcy in cardiomyocytes, varying from induction of reversible flip-flop of the plasma membrane phospholipids, to apoptosis and necrosis.

A new array of ternary lanthanide complexes of the form [Ln(hfa)(tpy-HImzphen)] have been synthesized and thoroughly characterized wherein Ln = La (), Eu (), Sm (), and Tb (); hfa = hexafluoroacetylacetonate; and tpy-HImzphen = 2-(4-[2,2':6',2″]terpyridin-4'-yl-phenyl)-1H-phenanthro[9,10-]imidazole. Incorporation of tpy-HImzphen onto the Ln-hfa moiety induced a bathochromic shift of the absorption window of the complexes into the visible region. Extensive investigations of the luminescence characteristics have been conducted both at RT and at 77 K to understand the deactivation pathways of the complexes.

For confirmation and/or screening purposes, rapid, selective, and precise chromatographic methods are required. In this vein, the utility of SiH columns (C18, UDC Cholesterol, and Diamond Hydride) with photodiode array UV absorption and single quadrupole MS detection for multi-modal separation of representative drugs from different drug classes on a single column using the same solvent reservoirs was investigated. For a conventional two column approach employing a combination of conventional C18 and silica columns operating in both the reversed phase chromatographic and hydrophilic interaction chromatographic modes, gradient analysis is required for the first column and there is a lack of retention on the second column for non-amine analytes.

Neural network and decision tree-based soft computing techniques are implemented in this work for the thorough analysis of the multichannel anion-sensing characteristics of an Os(II)-bipyridine complex derived from imidazole-4,5-bis(benzimidazole) ligand. With the aid of three imidazole NH protons in its outer coordination sphere, a substantial change in the spectral response as well as Os/Os potential is made possible upon treating with anions of varying basicity. Initial hydrogen bonding between NH protons and anions and thereafter complete proton transfer from the complex backbone probably take place in the process.

The oral absorption of exenatide, a type 2 diabetes medication, can be increased by employing lipid nanocapsules (LNC). To increase mucus permeability and exenatide intestinal absorption, reverse micelle lipid nanocapsules (RM-LNC) were prepared and their surface was modified with DSPE-PEG-FA. The RM-LNC with surface modification of DSPE-PEG-FA (FA-RM-LNC) were able to target enterocytes and reduce mucus aggregation in the intestine.
