Affinity precipitation is a bioseparation technique where the affinity ligand is coupled to a stimuliresponsive polymer. Stimuli-responsive polymers show abrupt, yet reversible, phase transition (precipitation) in response to a small change in an environmental parameter. The corresponding ligand conjugates can be used to co-precipitate and thereby capture and isolate target molecules from complex solutions such as culture supernatants and cell lysates. The approach is compatible with a 'discardibles only' type of downstream process and can be scaled over several orders of magnitude. This report discusses the set-up and development of affinity precipitation procedures, the related instrumentation and scale up, as well as applications for the isolation of proteins and polynucleotides.

Crosslinked Enzyme Aggregates (CLEAs) are favored for their operational stability and recyclability. However, the traditional CLEAs preparation may distort the enzyme's active site and reduce activity. Therefore, we developed a universally applicable crosslinked SpyCatcher scaffold system designed for the facile preparation of CLEAs.

This study optimized a one-step precipitation process for manganese recovery from a complex medium-bioleachate obtained from electric arc furnace dust (EAFD). The effects of pH variations and different precipitation agents, including acetone, ethanol, oxalic acid, and ammonium hydroxide, were investigated for manganese recovery. While acetone and ethanol facilitated precipitation, they did not lead to the formation of a specific manganese precipitate.

Naturally widespread ferrihydrite is unstable and often coexists with complex ions, such as the heavy metal ion Pb(II). Ferrihydrite could fix Pb(II) by precipitation and hydroxyl adsorption, but release Pb(II) with mineral aging. Gallic acid plays an important role in influencing the geochemical behavior of ferrihydrite-Pb, and anoxia is one of the factors influencing the transformation of mineral.

The death and clearance of nurse cells is a consequential milestone in Drosophila melanogaster oogenesis. In preparation for oviposition, the germline-derived nurse cells bequeath to the developing oocyte all their cytoplasmic contents and undergo programmed cell death. The death of the nurse cells is controlled non-autonomously and is precipitated by epithelial follicle cells of somatic origin acquiring a squamous morphology and acidifying the nurse cells externally.

Salivary extracellular vesicles isolation methods impact the robustness of downstream biomarkers detection.

Extracellular vesicles (EVs), crucial mediators in cell-to-cell communication, are implicated in both homeostatic and pathological processes. Their detectability in easily accessible peripheral fluids like saliva positions them as promising candidates for non-invasive biomarker discovery. However, the lack of standardized methods for salivary EVs isolation greatly limits our ability to study them.
