Presentation of (glyco)peptides by the class II major histocompatibility complex molecule Aq to T cells plays a central role in collagen-induced arthritis, an animal model for the autoimmune disease rheumatoid arthritis. A peptide library was designed using statistical molecular design in amino acid space in which five positions in the minimal mouse collagen type II binding epitope CII260-267 were varied. A substantially reduced peptide library of 24 peptides with diverse and representative molecular characteristics was selected, synthesized, and evaluated for the binding strength to Aq. A multivariate QSAR model was established by correlating calculated descriptors, compressed to its principle properties, with the binding data using partial least-square regression. The model was successfully validated by an external test set. Interpretation of the model provided a molecular property binding motif for peptides interacting with Aq. The information may be useful in future research directed toward new treatments of rheumatoid arthritis.

Exercise activates autophagy and lysosome system in skeletal muscle, which are known to play an important role in metabolic adaptation. However, the mechanism of exercise-activated autophagy and lysosome system in obese insulin resistance remains covert. In this study, we investigated the role of exercise-induced activation of autophagy and lysosome system in improving glucose metabolism of skeletal muscle.

Class I major histocompatibility complex (MHC-I) proteins play a pivotal role in adaptive immunity by displaying epitopic peptides to CD8+ T cells. The chaperones tapasin and TAPBPR promote the selection of immunogenic antigens from a large pool of intracellular peptides. Interactions of chaperoned MHC-I molecules with incoming peptides are transient in nature, and as a result, the precise antigen proofreading mechanism remains elusive.

Background: Understanding the genetic etiology of Alzheimer's disease (AD) has been a major focus of research in neurodegenerative diseases. Amid the three common allelic variants of the apolipoprotein E (APOE) gene in humans, called APOE ε2, ε3 and ε4, the ε4 allele is the most common genetic risk factor for late-onset AD, being found in 20% of the world population.

Method: We used Event-Related Potentials (ERP) and Event-Related Spectral Perturbation (ERSP) as features for classification of apolipoprotein E ϵ4 (APOE ε4) allele carriers in AD patients and healthy controls.

Background: While numerous blood biomarkers have been proposed for Alzheimer's disease (AD), only a few have demonstrated definitive diagnostic value. Recently, a set of phosphorylated Tau proteins, particularly pT217, have emerged as promising candidates with superior diagnostic performance. Given the development of pT217 antibodies by major global pharmaceutical companies, our goal is to create the best-in-class pT217 antibody, establishing it as the gold standard for diagnostics.

Background: The COVID-19 pandemic has significantly altered the etiological spectrum and epidemiological characteristics of pediatric respiratory diseases, and a profound understanding of these changes is crucial for guiding clinical treatment. The purpose of this study is to analyze the etiological patterns and epidemiological features of pathogens in bronchoalveolar lavage fluid (BALF) from children with pediatric lower respiratory tract infections (LRTIs) after the COVID-19 pandemic, with the aim of providing effective therapeutic evidence for clinical practice.

Methods: This study enrolled pediatric patients diagnosed with LRTIs who were treated and underwent BALF pathogen detection at our hospital from June 1, 2023, to June 1, 2024.
