Evaluation of mutagenic activity in supply water at three sites in the state of Rio Grande do Sul, Brazil.

In this study, the Salmonella/microsome assay, using the micro-suspension method, was utilized to evaluate water for public supply at three sites in Rio Grande do Sul. The first site selected was in an area under industrial influence and the others were in non-industrial reference areas. Based on 40 L samples of raw water and/or after conventional treatment, compounds were extracted with XAD4 resins using natural and acidic pH, and the extracts were analyzed in the TA98 and TA100 strains with and without S9. Raw water extracts in the industrial region induced 27.4 revertants/L (rev/L) for TA100+S9 up to 226.3 rev/L for TA100-S9, both for acidic pH extracts. After conventional treatment the responses varied from 20.6 rev/L (TA98-S9) for natural pH extracts to 755.5 rev/L (TA98-S9) for acidic pH extracts. For acidic extracts obtained from reference site samples, the response, with metabolic activation only, ranged from negative to minimal. Direct mutagenic responses in acidic extracts may be elevated in treated extracts, compared with raw water extracts, influenced by the presence of by-products of the chlorination process. However, the mutagenicity observed in the treated water extracts in the industrial area increased and reflected mainly the combination of directly and indirectly acting compounds in the source waters, that are heavily influenced by anthropogenic factors.

The effect of solvents with different polarities on the recovery of phytochemicals (carotenoids, capsaicinoids, and phenolic compounds) from habanero pepper (Capsicum chinense) and their association with antioxidant activity (ABTS and DPPH) was evaluated through Ultra-Performance-Liquid Chromatography coupled with a Photodiode Array Detector and a Electrospray Ionization Mass Spectrometry (UPLC-PDA-ESI-MS)-based chemometric analysis, including linear correlation, multiple linear regression, and principal component analysis (PCA). The solvent polarity scale was established according to solvent dielectric constants (ɛ). Color variation (ΔE) was used to determine the presence of carotenoids, with the highest ΔE obtained using low-polarity solvents (hexane and ethyl acetate).

A novel magnetic mesoporous fluorinated metal-organic framework material (FeO@MIP-206-F) has been synthesized specifically for application as an adsorbent for perfluoroalkyl carboxylic acids (PFCAs) extraction by magnetic solid-phase extraction (MSPE). The carefully designed FeO@MIP-206-F material features an appropriate porosity, open metal sites of Zr, and functional groups (fluorine and amino) conducive to the adsorption process. Novel and simple spectrophotometric and distance based procedures for thiols (L-cysteine, N-acetylcysteine, and glutathione) determination in biological fluids and pharmaceuticals have been proposed based on their inhibitory action on the oxidation of catechol in the presence of Agaricus bisporus crude extract (ABE). The influence of L-glycine, L-alanine, L-proline, L-methionine, L-cystine, ascorbic acid, uric acid, and bilirubin on the thiol determination has been investigated. Uric acid, bilirubin, L-cystine (oxidized thiol), and L-amino acids do not interfere with the determination.

A series of ten topiramate-phenolic acid conjugates (T1-T10) were synthesized, and evaluated for their pancreatic lipase inhibitory and antioxidant potentials. The design of the compounds reflected the structural attributes extracted from robust QSAR models developed for predicting the pancreatic lipase inhibition potency. Conjugate T4 competitively inhibited pancreatic lipase with IC value of 8.
