NG-497 Alleviates Microglia-Mediated Neuroinflammation in a MTNR1A-Dependent Manner.

Inflammation

January 2025

Department of Neurology, Nanjing Drum Tower Hospital, Affiliated Hospital of Medical School, Nanjing University, Nanjing, China.

Microglia-mediated neuroinflammation plays a crucial role in multiple neurological diseases. We have previously found that Atglistatin, the mouse Adipose Triglyceride Lipase (ATGL) inhibitor, could promote lipid droplets (LDs) accumulation and suppress LPS-induced neuroinflammation in mouse microglia. However, Atglistatin was species-selective, which limited its use in clinical settings.

Clinical Manifestations.

Alzheimers Dement

December 2024

University of California San Francisco, San Francisco, CA, USA.

Those with neurodegenerative conditions have an increased risk of developing delirium and there is some evidence that delirium may be a risk factor for neurodegenerative disorders. Similar to the interactions between sleep and neurodegenerative conditions, there is increasing evidence for bi-directional relationships between delirium and sleep disorders and disturbances. This presentation will provide an overview of the literature on those relationships, including discussion of the well-characterized changes in sleep that occur in individuals experiencing delirium.

Background: Variations in the circadian rhythm in older adults can negatively affect brain and cardiovascular health. As a consequence, poor cognitive functioning, an increase in the inflammatory response, alterations in sleep patterns, changes in mood and a decrease in motivation, energy and initiative are observed.

Method: We retrospectively examined the initial and final neuropsychological assessment of 250 MCI outpatients, 125 of whom had received daily 3-9 mg of a fast-release melatonin preparation p.

Insomnia in older adults: A review of treatment options.

Cleve Clin J Med

January 2025

Medical Director, Hartford Behavioral Health, Hartford, CT; Community Faculty, University of Connecticut, Farmington, CT.

Insomnia is a common and challenging complaint in older adults (> 65 years) because of age-related alterations in sleep physiology. Cognitive behavioral therapy for insomnia is the gold standard for treatment of insomnia in young as well as older patients. Both clinicians and patients often prefer the simplicity of medication, but risks associated with some hypnotics increase with age.

Combination of Apigenin and Melatonin with nanostructured lipid carriers as anti-inflammatory ocular treatment.

Int J Pharm

December 2024

Department of Pharmacy, Pharmaceutical Technology and Physical Chemistry, University of Barcelona, 08028 Barcelona, Spain; Institute of Nanoscience and Nanotechnology (IN(2)UB), University of Barcelona, 08028 Barcelona, Spain. Electronic address:

Ocular inflammation is a complex pathology with limited treatment options. While traditional therapies have side effects, novel approaches, such as natural compounds like Apigenin (APG) and Melatonin (MEL) offer promising solutions. APG and MEL, in combination with nanostructured lipid carriers (NLC), may provide a synergistic effect in treating ocular inflammation, potentially improving patient outcomes and reducing adverse effects.
