G protein-coupled receptors (GPCRs) are key players in cell communication. Several classes of such receptors have been identified. Although all GPCRs possess a heptahelical domain directly activating G proteins, important structural and sequence differences within receptors from different classes suggested distinct activation mechanisms. Here we show that highly conserved charged residues likely involved in an interaction network between transmembrane domains (TM) 3 and 6 at the cytoplasmic side of class C GPCRs are critical for activation of the gamma-aminobutyric acid type B receptor. Indeed, the loss of function resulting from the mutation of the conserved lysine residue into aspartate or glutamate in the TM3 of gamma-aminobutyric acid type B(2) can be partly rescued by mutating the conserved acidic residue of TM6 into either lysine or arginine. In addition, mutation of the conserved lysine into an acidic residue leads to a nonfunctional receptor that displays a high agonist affinity. This is reminiscent of a similar ionic network that constitutes a lock stabilizing the inactive state of many class A rhodopsin-like GPCRs. These data reveal that despite their original structure, class C GPCRs share with class A receptors at least some common structural feature controlling G protein activation.

The geochemical and chemical constituents of river water quality could be influenced by human activities and organic processes like water interacting with the lithogenic structure that the river flows through. Evaluating evidence based primary root of the predominant pollutant ions, their interactions as well as the factors controlling their dominance is crucial in studies regarding water environment and hydrology especially as most studies focus on theoretical methods. In order to understand the water cycle, safeguard surface water resources, and preserve the human environment, this study evaluated surface water hydro-chemical facies, quality dynamics, and portability in southern Nigeria using multivariate statistical approaches by analyzing selected hydro-chemical characteristics as indicators of pollution along the river during wet and dry seasons.

The faithful charging of amino acids to cognate tRNAs by aminoacyl-tRNA synthetases (AARSs) determines the fidelity of protein translation. Isoleucyl-tRNA synthetase (IleRS) distinguishes tRNA from tRNA solely based on the nucleotide at wobble position (N34), and a single substitution at N34 could exchange the aminoacylation specificity between two tRNAs. Irrespective of the specific see-through device, obtaining optimal transparency remains the primary goal. In this work, we introduce a general strategy to enhance the transparency of various see-through devices. We achieve this by structuring the colored functional materials into imperceptible three-dimensional mesh lines, addressing a common challenge in multi-layer structures where each layer causes a reduction in transparency due to their color or opacity.
