Previous studies suggest that chemokine CXCL12 and its receptor CXCR4 play important roles in embryonic development, stem cell trafficking, and inflammatory reactions. It is a central factor in directing cell movement of many physiologic and pathologic processes. The mechanisms of cancer cell metastasis and white blood cell trafficking are similar. CXCL12-CXCR4 axis exerts important effects in regulating growth, invasion, metastasis, and secretion of malignant cells. Data from animal experiments suggest that CXCR4 may be an important therapeutic target in inhibiting malignant growth and metastatic behavior of tumor cells. This review focused on the role of CXCL12-CXCR4 axis in regulating tumor metastasis and progression, and the molecular mechanisms that are essential to this process.

These cells migrate to and proliferate in the bone marrow and lymphoid tissues. CXCL12 and CXCR4 proteins and mRNAs were monitored in the dorsal root ganglia (DRGs) of lumbar (L4-L5) and cervical (C7-C8) spinal segments of naïve rats, rats subjected to sham operation, and those undergoing unilateral complete sciatic nerve transection (CSNT) on post-operation day 7 (POD7). Immunohistochemical, Western blot, and RT-PCR analyses revealed bilaterally increased levels of CXCR4 protein and mRNA in both lumbar and cervical DRG neurons after CSNT. Similarly, CXCL12 protein levels increased, and CXCL12 mRNA was upregulated primarily in lumbar DRGs ipsilateral to the nerve lesion.

We previously demonstrated that C-X-C Motif Chemokine Ligand 12 (CXCL12) is primarily secreted by dermal fibroblasts in response to androgens and induces hair miniaturization in the mouse androgenic alopecia (AGA) model. However, the direct effects of androgen-induced CXCL12 on dermal papilla cells (DPCs) and dermal sheath cup cells (DSCs) have not been demonstrated. First, we compared single-cell RNA sequencing data between mouse and human skin, and the results show that CXCL12 is highly co-expressed with the androgen receptor (AR) in the DPCs and DSCs of only human hair.

Neutrophils, particularly low-density neutrophils (LDNs), are believed to contribute to acute COVID-19 severity. Here, we showed that neutrophilia can be detected acutely and even months after SARS-CoV-2 infection in patients and mice, while neutrophil depletion reduced disease severity in mice. A key factor in neutrophilia and severe disease in infected mice was traced to the chemokine CXCL12 secreted by bone marrow cells and unexpectedly, endothelial cells.
