Unlabelled: Cyclosporin A (CyA), a potent immunosuppressant, was used to determine the hepatotoxic effect in long-term treatments. Male Wistar rats were used in these experiments. They were given CyA chronically at doses used in patients for 120 days, and at doses of 5, 10, 15 and 20 mg kg(-1) day(-1). These doses amount to CyA values in blood of 200 +/- 24, 314 +/- 40, 445 +/- 33 and 598 +/- 53 ng ml(-1), respectively. A significant increase in glutamate dehydrogenase (GLDH) was found in the groups treated with 15 and 20 mg kg(-1) day(-1), which would point to mitochondria as the potential target of the toxic action of CyA. The mitochondrial respiratory chain of rat livers was studied in enzyme complexes I and II. Enzyme complex I was determined by spectrophotometry at 340 nm using NADH oxidase with the respirable substrate 10 mm NADH; enzyme complex II was determined by monitoring succinate dehydrogenase by oxymetry using the respirable substrate 10 mm succinate. The results show the inhibition of NADH oxidase in the groups treated with 10, 15 and 20 mg kg(-1) day(-1), an effect dependent both on time and on CyA concentration. Enzyme complex II showed a decrease in oxygen consumption. These findings were confirmed by histological studies (hematoxylin-eosin technique).

Conclusions: Long-term treatment with CyA at doses of 15 and 20 mg kg(-1) day(-1), amounting to concentrations in blood of 445 +/- 33 and 598 +/- 53 ng ml(-1), causes alterations in the mitochondria, revealed by the increase in serum GLDH and by the functional alteration of enzyme complexes I and II of the mitochondrial respiratory chain.

Two-dimensional (2D) covalent organic frameworks (COFs) with designable pore structures can be synthesized under the guidance of topology diagrams. Among the five existing edge-transitive topological nets, topology is considered a fine candidate for constructing COFs with ultramicropores. However, all of the reported COFs with topology need the use of -symmetric monomers, which are limited in compound type and difficult to synthesize.

Background And Purpose: Eukaryotic elongation factor 2 kinase (eEF2K) belongs to the Ca/calmodulin-dependent protein kinase family. We previously revealed that A484954, a selective eEF2K inhibitor, caused hypotensive and diuretic effects via the production of nitric oxide (NO) in spontaneously hypertensive rats. Otsuka Long-Evans Tokushima Fatty (OLETF) rats are hypertensive because of obesity and type 2 diabetes.

Parasitic infestations are one of the most economically important disease conditions in the Indian major carps including mrigal, Cirrhinus mrigala. This study reported the biosafety and tissue withdrawal of in-feed administered antiparasitic drug, emamectin benzoate (EMB). To evaluate the biosafety of the drug, behaviour, growth and tissue changes in Cirrhinus mrigala was recorded the following in-feed administration of EMB up to 10 times (T1-50 μg kg fish day (1×), T2-125 μg kg fish day (2.

Tiered intake assessment for propylene glycol in beverages.

Food Addit Contam Part A Chem Anal Control Expo Risk Assess

January 2025

American Beverage Association, Washington, DC, USA.

A tiered intake assessment approach is presented and applied to derive estimates of maximum potential global propylene glycol (PG) intake from beverage sources. The US and UK markets served as surrogates for the world and the EU region, respectively, to determine the maximum potential exposure for PG in various subpopulations, including brand-loyal consumers. Conservative intake estimates for PG used in non-alcoholic beverages were calculated for toddlers less than 3 years (y), young children 3-9 y, adolescents 10-17 y, adults 18-64 y, elderly 65-74 y, very elderly 75+ y, based on assumed uses in high beverage consumption markets, leveraging either the 2-day food consumption data from the 2013-2016 US National Health and Nutrition Examination Survey or the 4-day food consumption data from the 2008-2017 UK National Diet and Nutrition Survey Rolling Programme.
