Methoxy poly(ethylene glycol)-b-poly(caprolactone) (MePEG-b-PCL) copolymers with varying PEG block lengths and a constant PCL block length were synthesized by cationic ring-opening polymerization and used to form nano-sized micelles. Due to their small size and superior in vitro stability, the MePEG(5000)-b-PCL(5000) micelles were selected for further in vitro characterization and an in vivo evaluation of their fate and stability following intravenous (i.v.) administration. Specifically, (3)H-labelled MePEG(5000)-b-PCL(5000) micelles were i.v. administered to Balb/C mice at copolymer doses of 250, 2 and 0.2 mg/kg in order to examine the distribution kinetics of (1) copolymer assembled as thermodynamically stable micelles, (2) copolymer assembled as thermodynamically unstable micelles and (3) copolymer unimers, respectively. Overall, it was found that when the copolymer is assembled as thermodynamically stable micelles the material is effectively restricted to the plasma compartment. Interestingly, the copolymer was found to have a relatively long circulation half-life even when administered at a dose that would likely fall to concentrations below the CMC following distribution. Analysis of plasma samples from this group revealed that even 24 h post-administration a significant portion of the copolymer remained assembled as intact micelles. In this way, this study demonstrates that the hydrophobic and semi-crystalline nature of the PCL core imparts a high degree of kinetic stability to this micelle system.

Due to the straightforward single-step synthesis, amphiphilic gradient copoly(2-oxazoline)s are becoming more popular alternative to their block analogue for the development of next-generation drug delivery systems. Here, we investigated the influence of polymer architecture on the physiochemical and biological assessment of nanoformulations formed by the self-assembly of gradient copoly(2-oxazoline)s. Two different architectures were synthesized: hydrophilic-grad-hydrophobic (mono-gradient) and hydrophobic-grad-hydrophilic-grad-hydrophobic (di-gradient) which contained a hydrophilic monomer, 2-ethyl-2-oxazoline (EtOx) and a hydrophobic monomer, 2-phenyl-2-oxazoline (PhOx).

This study aims to explore the development of natural bio-based amphiphilic block copolymers for drug delivery applications. We investigated block copolymers derived from tamarind seed xyloglucan and solanesol, focusing on their synthesis, structural analysis, aqueous self-assembly, and drug encapsulation. Specifically, xyloglucan hydrolysate segments with number-average degrees of polymerization (DPs) of between 8 and 44 (XOS, XMS, XMS, XMS, and XMS) were used as the hydrophilic blocks, whereas plant-sourced solanesol was selected as the hydrophobic segment.

In this study, we applied a systematic approach to establish an iterative workflow and to drive the chemical design of thermosensitive, in situ forming injectables as a function of the intended target product profile. Self-assembly, mechanical properties, physical state, and thermal transition behavior were assessed via nuclear magnetic resonance, oscillatory rheology, turbidimetry and visual inspection techniques. Thus, poly(N-isopropylacrylamide) (PNIPAM) and poly(2-alkyl-2-oxazoline)s (PAOx)s with LCSTs below body temperature were studied before and after grafting them onto azido-substituted hyaluronic acid (HA) via strain-promoted azide-alkyne cycloaddition (SPAAC).

Bottlebrush block copolymers (BBCPs) are a unique class of materials that contain a backbone with densely grafted and chemically distinct polymeric side chains. The nonlinear architecture of BBCPs provides numerous degrees of freedom in their preparation, including control over key parameters such as grafting density, side chain length, block arrangement, and overall molecular weight. This uniquely branched structure provides BBCPs with several important distinctions from their linear counterparts, including sterically induced side chain and backbone conformations, rapid and large self-assembled nanostructures, and reduced or eliminated entanglement effects (assuming sufficient grafting density and that the molecular weight of the side chains is below their respective entanglement molecular weight).
