Establishment of facultative sexuals.

The existence of sex is one of the major unsolved problems in biology. We use computer simulations to model conditions in which sex may first become established. We develop an individual-based population model and show that a hypothetical facultative sex gene can fix, provided that the initial cost is low. It is demonstrated that the equilibrium fitness in the population increases with increasing population size and decreasing mutation rate. The probability of the establishment of the sex gene is found not to be directly related to the fitness difference between the asexual and sexual populations. This change in fitness on changing the parameters of the model is investigated.

Reproductive success and ultimately species survival at a population level is contingent on a plethora of neuroendocrine signals working in concert to regulate gonadal function and reproductive behavior. Among these, the neuropeptide kisspeptin (encoded by the KISS1/Kiss1 gene) has emerged as the master regulator of the hypothalamic-pituitary-gonadal axis. Besides the hypothalamus, both kisspeptin and its cognate receptor are extensively expressed throughout cortico-limbic brain structures in rodents and humans, which are regions traditionally implicated in behavioral and emotional responses.

Imprinting abnormalities pose a significant challenge in applications involving embryonic stem cells, induced pluripotent stem cells, and animal cloning, with no universal correction method owing to their complexity and stochastic nature. In this study, we targeted these defects at their source-embryos from same-sex parents-aiming to establish a stable, maintainable imprinting pattern de novo in mammalian cells. Using bi-paternal mouse embryos, which exhibit severe imprinting defects and are typically non-viable, we introduced frameshift mutations, gene deletions, and regulatory edits at 20 key imprinted loci, ultimately achieving the development of fully adult animals, albeit with a relatively low survival rate.
