In the present study, the effects of long-term pinealectomy on tryptophan, 5-hydroxytryptamine (5-HT or serotonin), 5-hydroxy-3-indoleacetic acid (5-HIAA), and tryptophan hydroxylase and monoamine oxidase activities were studied in preoptic area-anterior hypothalamus (POA-AH) and in the medial and posterior hypothalamus of the rat. After pinealectomy, 5-HT levels decreased significantly in medial hypothalamus but increased in the POA-AH. The levels of 5-HIAA decreased significantly in the POA-AH and medial hypothalamus. Tryptophan levels remained unchanged while tryptophan hydroxylase activity diminished significantly in POA-AH and medial hypothalamus. Monoamine oxidase activity remained unchanged in the hypothalamic regions. These results suggest that pinealectomy induces differential inhibitory actions on the serotoninergic terminal regions, mainly in anterior and medial hypothalamic areas.

Objective: To explore the effects of aging on the levels of reproduction-related mRNA genes including Gnrh, KISS1/KISS1r, estrogen receptor-alpha (ERα), estrogen receptor-beta (ERβ) and progesterone receptor (PR) in hypothalamus.
Methods: Proestrus and metestrus in young (3-4 months) and middle-aged (10-11 months) female mice and diestrus in senile (18-19 months) female mice were observed. And the levels of related mRNA genes in preoptic area anterior hypothalamus (POA-AH) and medial basal hypothalamus (MBH) were determined by real-time polymerase chain reaction (RT-PCR).

Background/aims: Adult mice lacking functional GABAB receptors (GABAB1KO) show altered Gnrh1 and Gad1 expressions in the preoptic area-anterior hypothalamus (POA-AH) and females display disruption of cyclicity and fertility. Here we addressed whether sexual differentiation of the brain and the proper wiring of the GnRH and kisspeptin systems were already disturbed in postnatal day 4 (PND4) GABAB1KO mice.
Methods: PND4 wild-type (WT) and GABAB1KO mice of both sexes were sacrificed; tissues were collected to determine mRNA expression (qPCR), amino acids (HPLC), and hormones (RIA and/or IHC).

Previous studies showed that bilateral lesions of the male ferret's preoptic area/anterior hypothalamus (POA/AH), centered in the sexually dimorphic nuclei present in this region, caused subjects to seek out a same-sex male, as opposed to a female conspecific. Male subjects with POA/AH lesions (which were also castrated and given estradiol) displayed female-typical receptive behavior in response to neck gripping by a stimulus male, implying that subjects' approaches to a same-sex conspecific were sexually motivated. We asked whether the effect of POA/AH lesions on males' partner preference reflects a shift in the central processing of body odorant cues so that males come to display a female-typical preference to approach male body odorants.
