Replicative double-stranded RNA (dsRNA) is useful in preliminary identification of Cucumber mosaic virus and its satellite RNA (satRNA). This plant pathogen complex yields sufficient quantity of the replicative RNA form that can be isolated by chromatography on chemically unmodified graded cellulose powder (CF-11). In this work, much faster and more efficient procedure using DEAE monoliths was developed in which dsRNA was separated from other species in total nucleic acids extract originating from the infected plant tissue. The developed chromatographic method revealed the pathogens' presence in only 15 min, avoiding nucleic acid precipitation and electrophoretic analysis.

Engineered transfer RNA is an emerging therapeutic modality, particularly suited to treatment of diseases caused by genetic disorders based on premature termination codons, frameshifts, or missense mutations. It is also extensively used in reprogramming of translation systems to generate non-canonical amino acid-containing proteins and peptides, such as in mRNA display. Cellulose monolith has garnered significant interest in the field of biochromatography, which lies in its interconnected porous structure, large surface area and biocompatibility. In this context, we propose a novel approach for preparing cellulose monoliths using an aqueous two-phase system devoid of any organic solvents and surfactants. In this strategy, emulsifying cellulose solution into PEG 20,000 solution gives bicontinuous aqueous phases and further porous cellulose monolith after regeneration of dissolved cellulose.

Elution of strong and weak anion exchangers with sodium chloride gradients is commonly employed for analysis of sample mixtures containing different isomers of plasmid DNA. Gradient elution of a weak anion exchanger (diethylaminoethyl) in the presence of guanidine hydrochloride (Gdn) roughly doubles resolution between open-circular (oc) and supercoiled (sc) isomers.
