In the work it was shown that fungal infection of insects increased the total esterase and glutathione-S-transferase activities of hemolymph. Acid and alkaline phosphatase activity in infected larvae remained at the same level as in intact larvae. Fungi-infected bee pyralid larvae turned out to be 1.46 times more resistant to phosphoorganic insecticide malation as compared to intact larvae. On the other hand, treating insects with the inhibitors of detoxicating enzymes caused a sharp decrease in the insects' resistance to entomophathogenic fungi. The work discusses possible participation of detoxicating enzymes in the formation of the insects' resistance to entomopathogenic fungi as well as new strategies of the development and creation of complex biopreparations based on entomopathogenic microorganisms and inhibitors of detoxicating enzymes.

Cadmium accumulation potential and detoxication mechanism of Koenigia tortuosa: A novel extremely hardy plant from high altitude lead-zinc mine in Qinghai-Tibet Plateau.

State Key Laboratory of Geohazard Prevention and Geoenvironment Protection, College of Ecology and Environment, Chengdu University of Technology, Chengdu, 610059, PR China.

Hardy plants play a crucial role in restoring high-altitude tailings ponds, but the accumulation of potentially toxic elements (PTEs) and detoxification mechanisms in alpine plants are understudied. This study first investigated the cadmium (Cd) accumulation capacity and detoxification mechanisms by comparative transcriptomics with different Cd stress (0, 5, 10, 20 and 40 mg L Cd) of Koenigia tortuosa from a lead-zinc mine (4950 m above sea level) in Qinghai-Tibet Plateau. The findings revealed that, despite elevated Cd concentrations suppressed the growth of Koenigia tortuosa, the plant retained a notable ability to accumulate Cd.

Glutathione S-transferases (GSTs) are evolutionarily conserved enzymes crucial for cell detoxication. They are viewed as having evolved in cyanobacteria, the ancient photosynthetic prokaryotes that colonize our planet and play a crucial role for its biosphere. Xi-class GSTs, characterized by their specific glutathionyl-hydroquinone reductase activity, have been observed in prokaryotes, fungi and plants, but have not yet been studied in cyanobacteria.

Nanozymes, a category of nanomaterials with exceptional enzyme-like activity, exhibit the significant promise to overcome the inherent limitations of natural enzymes. Inspired by the active site structure of natural laccase, a biomimetic MA-Cu nanozyme with three-dimensional network structure was constructed in water system through one-step complexation based on the specific coordination between nitrogen-rich triazine heterocyclic melamine and Cu, in a facile, green and economical manner. Compared to natural laccase, MA-Cu possesses superior multi-enzyme mimicking activity, stability and cost-effectiveness.

Mutations Selectively Evolving Peroxidase Activity Among Alternative Catalytic Functions of Human Glutathione Transferase P1-1.

Antioxidants (Basel)

November 2024

Department of Biochemistry and Biophysics, Arrhenius Laboratories, Stockholm University, SE-10691 Stockholm, Sweden.

Glutathione transferases are detoxication enzymes with broad catalytic diversity, and small alterations to the protein's primary structure can have considerable effects on the enzyme's substrate selectivity profile. We demonstrate that two point mutations in glutathione transferase P1-1 suffice to generate 20-fold enhanced non-selenium-dependent peroxidase activity indicating a facile evolutionary trajectory. Designed mutant libraries of the enzyme were screened for catalytic activities with alternative substrates representing four divergent chemistries.
