The cell counting of colonial Microcystis spp. is a rather difficult and error-prone proposition, as this genus forms irregularly-shaped and irregularly-sized colonies, which are packed with cells. Thus, in order to facilitate a cell count, four methods of dividing the colonies into single cells were compared, including vortexing, sonication, TiO2 treatment, and boiling. As a result, the boiling method was determined to generate the greatest number of single cells from a colony, and all colonies were found to have divided completely after only 6 min of treatment. Furthermore, no significant cell destruction, which might alter the actual cell density, was detected in conjunction with the boiling method (P = 0.158). In order to compute the cell number more simply, the relationship between the colony size and the cell number was determined, via the boiling method. The colony volume, rather than the area or diameter was correlated more closely with the cell number (r2 = 0.727), thereby suggesting that the cell numbers of colonial Microcystis sp. can also be estimated effectively from their volumes.

Continuous microenvironment modulation is an ongoing challenge in wound dressing, which includes excessive exudate absorption, oxygen delivery, bacterial inhibition and angiogenesis. Herein, we developed an construction strategy to fabricate a self-retaining double-layered wound dressing, where the top layer precursor was composed of Ca-containing polyvinyl butyral (PVB) solution dispersed with hydroxypropyl methylcellulose (HPMC) particles, and the bottom one consisted of sodium alginate (Alg) solution blended with Ag-doped mesoporous bioactive glass powders (Ag-MBG). When in use, both precursors were simultaneously squeezed out from the twin nozzles connected to the individual chambers of a twin-chambered syringe, whereby Ca in the top layer rapidly migrated downwards to crosslink Alg in the bottom layer, leading to the formation of an Alg/Ag-MBG (AA) functional hydrogel for filling an irregular wound.

Soxhlet extraction is a method recommended by the Association of Official Analytical Chemists (AOAC) to determine the lipid content in plant samples. Generally, n-hexane (toxicity grade 5) is used as the solvent (≈300 mL; ≈30 g sample) at boiling temperatures (69 °C) for long times (≤16 h) under a chilled water reflux (≈90 L/h), proportionally aggravated by the number of repetitions and samples determined. In this sense, the technique is neither safe nor sustainable for the analyst or the environment.
