Monolithic media have found widespread use as excellent tools for fast analytical separations of small molecules, proteins, pDNA and viruses. Polymethacrylate monoliths with large channels are attractive for capturing large molecules, like immunoglobulins, DNA, and viruses. For preparative purposes, these monoliths are operated in radial flow mode. Band spreading in monoliths is extremely low and mostly dominated by the contribution of extra column effects. The model used here had a single axial dispersion coefficient which lumps together extra column effects and the intrinsic band spreading of the monolithic material to characterize the adsorption of proteins and pDNA on polymethacrylate ion-exchange monoliths. Due to the fact that the performance of the monolith was unaffected by the velocity within the applied range, and due to highly favourable adsorption isotherms, a constant pattern model could be applied to predict preparative runs on radial flow units assuming axial flow for modelling.

Background: Sample extraction is one of the most critical steps in most of the analytical processes. Nowadays, there is a demand for simple approaches that can effectively extract and concentrate target analytes from complex matrices, like biofluids, with accurate and reliable results.

This work relates to the preparation of novel and promising stationary phases containing inorganic-organic composites for capillary liquid and gas chromatography. A naturally occurring montmorillonite was introduced to polymethacrylate monoliths, then used under different conditions of GC and HPLC at the same time. A monolith of poly(methacrylic acid-co-ethylene glycol dimethacrylate) has been immobilised to a nitrocellulose strip by radical photopolymerisation to be used in the extraction of psychoactive substances in biological fluids. Codeine, methylone, amphetamine, methamphetamine, 3,4-methylenedioxymethamphetamine, butylone, norketamine, ketamine, heroin, cocaine, lysergic acid diethylamide and fentanyl were employed as model drugs and final extracts were analysed by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). Polymerisation parameters were adjusted in order to obtain a stable and homogeneous layer of monolith onto the nitrocellulose strip.

For the design of novel potent molecules against therapeutic protein targets produced in a low quantity or that are very expensive, the development of miniaturized analytical techniques is of crucial importance. One challenging target is the receptor binding domain (RBD) of the SARS-CoV-2-spike protein (S), which mediates the binding of the virus to host cells. In the present study, the RBD protein was thus immobilized on polymethacrylate monoliths prepared in a miniaturized capillary column (25 μm internal diameter; 70 mm length) by polymerization, which could offer low backpressure in Nano LC at 30 nL min.
