Bioassay-guided fractionation of the MeOH extract from the octocoral Muricea austera collected in the Pacific coast of Panama led to the isolation of eight compounds, including three tyramine derivatives (1-3), two steroidal pregnane glycosides (4, 5), and three sesquiterpenoids (6-8). Compounds 2-5 are new natural products, and their structures were determined on the basis of their spectroscopic data (HRMS, 1D and 2D NMR, and CD studies). The antiprotozoal activities of the natural compounds 1-8 as well as those of a series of synthetic glycosides (11-22) and tyramine derivatives (23-35) were evaluated in vitro against a drug-resistant Plasmodium falciparum and intracellular form of Trypanosoma cruzi.

Background: Coral-associated microbiomes vary greatly between colonies and localities with functional consequences on the host. However, the full extent of variability across the ranges of most coral species remains unknown, especially for corals living in deep waters which span greater ranges. Octocorals are important zoobenthic organisms, contributing to structural heterogeneity and species diversity on hardgrounds. Their persistence amidst global coral reef degradation and ocean acidification, has prompted renewed interest in this taxon. Octocoral assemblages at 52 sites in continental Ecuador and Galápagos (23 species, 3742 colonies) were examined for composition, size distributions within and among populations, and connectivity patterns based on ocean current models.

The genus is considered abundant and widely distributed along the eastern Pacific. Its occurrence in shallow waters has been recognised; however species from deeper than 30 m have been rarely recorded. During the 2005 R/V Urracá expedition along the north and central Pacific coast of Costa Rica several octocoral specimens were collected by bottom trawling from 30 to 150 m yielding new species and new records.

Octocorals are sources of novel but understudied microbial diversity. Conversely, scleractinian or reef-building coral microbiomes have been heavily examined in light of the threats of climate change. Muricea californica and Muricea fruticosa are two co-occurring species of gorgonian octocoral abundantly found in the kelp forests of southern California, and thus provide an excellent basis to determine if octocoral microbiomes are host specific.

The long-term goal of our study is to understand the internal organization of the octocoral stem canals, as well as their physiological and functional role in the growth of the colonies, and finally to assess the influence of climatic changes on this species. Here we focus on imaging tools, namely acquisition and processing of three-dimensional high-resolution images, with emphasis on automated extraction of canal pathways. Our aim was to evaluate the feasibility of the whole process, to point out and solve - if possible - technical problems related to the specimen conditioning, to determine the best acquisition parameters and to develop necessary image-processing algorithms.
