Use of stable isotopes to evaluate the functional effects of nutrients.

Purpose Of Review: Lifestyle, particularly dietary habits, plays a major role in the increasing prevalence of obesity, type 2 diabetes and atherosclerosis. Understanding how diet and specific foods can modify important functions of the body, in a beneficial or detrimental way, is therefore important. This review presents recent advances in the use of stable isotopes to investigate how nutrients can influence pathways of glucose, lipids and protein metabolism, and also intestinal absorption, body composition and fat mass turnover.

Recent Findings: Recent developments have focused mainly on the measurement of intratissular metabolic pathways, particularly in lipids and proteins. Advances in the field of cell proliferation rates, body composition and energy expenditure have also been made. Many of these developments rely on the use of deuterated water to trace multiple metabolic pathways.

Summary: Stable isotopes can now be used to measure the kinetics and oxidation rate of circulating metabolites and also body composition, colonic fermentation, intestinal absorption, and important intratissular metabolic pathways, such as gluconeogenesis, lipogenesis, cholesterol synthesis, the turnover rate of triglycerides, individual protein synthesis and cell proliferation. Deuterated water has emerged as a powerful and versatile tool, allowing the simultaneous investigation of several aspects of protein, lipid and glucose metabolism.

Iron not only influences the activity of anammox bacteria (AnAOB) but also participates in complex Fe-N cycles. In this study, the advanced N isotope tracing method was set up to quantify the potential rates of full nitrogen metabolic pathways under different ferrous iron (Fe and FeO) within two identical anammox granular reactors. The results indicated that both Fe and FeO enhanced AnAOB activity.

Single Cell-Pair Proteomics for Decoding Immune-Cancer Cell Interactions.

The efficacy of cancer immunotherapy is significantly influenced by the heterogeneity of individual tumors and immune responses. To investigate this phenomenon, a microfluidic platform is constructed for profiling immune-cancer cell interactions at the single-cell proteomics level for the first time. Based on the platform, a comprehensive workflow is proposed for achieving accurate single-cell pairing of an immune cell and a cancer cell with low cell damage and high success rate up to 95%, cell pair co-culture, and real-time microscopic monitoring of the cell-pair interactions, cell pair retrieval, mass spectrometry-based proteomic analysis of singe cell pairs, and decoupling of the proteomic information for each cell within the cell pair with the stable-isotope labeling method.

Naturally occurring bedded salt deposits are considered robust for the permanent disposal of heat-generating nuclear waste due to their unique physical and geological properties. The Brine Availability Test in Salt (BATS) is a US-DOE Office of Nuclear Energy funded project that uses heated borehole experiments underground (∼655 meters depth) at the Waste Isolation Pilot Plant (WIPP) in the bedded salt deposits of the Salado Formation to investigate the capacity for safe disposal of high-level, heat generating nuclear waste in salt. Uncertainties associated with brine mobility near heat-generating waste motivates the need to characterize the processes and sources of brine in salt deposits.
