So far, no antiangiogenic activity of wogonin, a flavonoid, on human umbilical vein endothelial cells (HUVECs) has been demonstrated. The aim of this study was to investigate the effects of wogonin on IL-6-induced angiogenesis in HUVEC cultures and chorioallantoic membrane (CAM) neovascularization. The in vivo CAM model was applied to evaluate the percentage of new vessel formations, followed by measurement of endothelial migration and tube formation in HUVEC cultures. The results revealed that wogonin (10(-8) approximately 10(-5)M) concentration-dependently inhibited IL-6-induced angiogenesis. The signaling pathway was through down-regulation of the autocrine loop of VEGF and VEGFR-1, but not of VEGFR-2. Furthermore, the regulating loop of the IL-6 receptor complex was also attenuated via expression of sIL-6Ralpha and gp130, but not of IL-6/IL-6R binding density. We conclude that wogonin is a suppressive agent of the autoregulated loop of VEGF, VEGFR-1 and the IL-6 receptor complex.

A molecular interaction between maternal endometrium and implanting conceptus can lead to activation of a variety of transcription factors that regulate expression of several genes necessary for the process of embryo implantation. While, signal transducer and activator of transcription 3 (STAT3) is responsible for decidualization and epithelial remodeling in humans and mice, its role in porcine endometrium has not been explored before. In the present study, we observed a pregnancy dependent increase in gene and protein expression of STAT3.

Psoriasis is a common chronic immune-mediated skin disease characterized by hyperproliferation and aberrant differentiation of keratinocytes and massive infiltration of inflammatory immune cells. Recent studies showed that Signal Transducer and Activator of Transcription 3 (STAT3), which plays an important role in cell survival, proliferation, differentiation, angiogenesis, and immune responses, is constitutively activated in epidermal keratinocytes of human psoriatic skin lesions. In addition, STAT3 promotes the differentiation and expansion of T cells and facilitates cytokine production, thereby exacerbating the condition of psoriasis.

Ca /calmodulin-dependent protein kinase II (CaMKII) is a ubiquitous serine threonine kinase with established roles in physiological and pathophysiological vascular remodeling. Based on our previous study demonstrating that CaMKIIδ promotes thrombin-induced endothelial permeability and recent reports that CaMKII may contribute to inflammatory remodeling in the heart, we investigated CaMKIIδ-dependent regulation of endothelial function downstream of an interleukin-6 (IL-6)/JAK/STAT3 signaling axis. Upon treatment with IL-6 and its soluble receptor (sIL-6r), CaMKIIδ expression is significantly induced in HUVEC.
