Aim: To prepare and characterize the monoclonal antibody (mAb) against sperm protein 17 (Sp17).
Methods: Human Sp17 cDNA was cloned and recombinant Sp17 protein was expressed as a fusion protein with an N-terminal 6-His tag. The purified recombinant Sp17 protein was used to immunize BALB/c mice for preparing mAb. mAb-produced hybridoma cells were screened by ELISA and the specificity of mAb was analyzed by immunohistochemical staining and blocking test.
Results: Two hybridoma cells (3C12 and 3D6) secreting mAb against Sp17 were developed. The isotypes of these two mAbs, 3C12 and 3D6, were IgG(1) and IgM, respectively. ELISA detection showed that titers of mAbs, 3C12 and 3D6, were 1:64, 1:32 in cultured supernatant and 1:1 x 10(5), 1:5 x 10(4) in ascites, respectively. The results of immunohistochemical staining and blocking test indicated that 2 mAb specifically bound to Sp17 in human and rat testis and human ejaculated spermatozoa. Anti-Sp17 mAb also detected Sp17 in ovarian cancer.
Conclusion: The successful preparation of anti-Sp17 mAb will be useful for assessing the native distribution and aberrant expression of Sp17 protein.

In the present investigation, we reported a label-free and highly effective immunosensor for the first time employing a nanostructured molybdenum disulfide nanosheets@reduced graphene oxide (nMoS NS@rGO) nanohybrid interface for the determination of sperm protein 17 (Sp17), an emerging cancer biomarker. We synthesized the nMoS NS@rGO nanohybrid using a one-step hydrothermal technique and then functionalized it with 3-aminopropyltriethoxysilane (APTES). Furthermore, the anti-Sp17 monoclonal antibodies were covalently attached to the APTES/nMoS NS@rGO/indium tin oxide (ITO) electrode utilizing 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide--hydroxy succinimide (EDC-NHS) coupling chemistry.

Herein, we report the results of the studies relating to developing a simple, sensitive, cost-effective, and disposable electrochemical-based label-free immunosensor for real-time detection of a new cancer biomarker, sperm protein-17 (SP17), in complex serum samples. An indium tin oxide (ITO) coated glass substrate modified with self-assembled monolayers (SAMs) of 3-glycidoxypropyltrimethoxysilane (GPTMS) was functionalized via covalent immobilization of monoclonal anti-SP17 antibodies using EDC(1-(3-(dimethylamine)-propyl)-3-ethylcarbodiimide hydrochloride) - NHS (N-hydroxy succinimide) chemistry. This study aims to evaluate the expression of genes associated with the fertilisation potential and embryo development, sperm DNA fragmentation (SDF), and acrosome reaction in male partners of infertile couples with different sperm parameters compared to fertile men. First, male partners of infertile couples with abnormal (N = 25) and normal sperm parameters (N = 25), and fertile men (N = 10) were included in experimental groups I, II, and controls respectively. The mRNA levels of the Annexin A2 (ANXA2), Sperm protein 17 (SP17), Plasma serine protease inhibitor (SERPINA5), and Peroxiredoxin-2 (PRDX2) genes and SDF were evaluated.
