Galactomannan hydrolysis results from the concerted action of microbial endo-mannanases, manosidases and alpha-galactosidases and is a mechanism of intrinsic biological importance. Here we report the identification of a gene cluster in the aerobic soil bacterium Cellvibrio mixtus encoding enzymes involved in the degradation of this polymeric substrate. The family 27 alpha-galactosidase, termed CmAga27A, preferentially hydrolyse galactose containing polysaccharides. In addition, we have characterized an enzyme with epimerase activity, which might be responsible for the conversion of mannose into glucose. The role of the identified enzymes in the hydrolysis of galactomannan by aerobic bacteria is discussed.

Whole-cell bacteria overexpressing a combo of enzymes capable of breaking down complex lignocellulosic components of cell wall is a path-breaking innovation that is eco-friendly for agricultural waste processing and sustainable environment. In this study, a whole-cell overexpressing the enzyme alpha-galactosidase is used to biodegrade sugarcane bagasse, presenting a sustainable approach for agricultural waste utilization. Alpha-galactosidase is an enzyme that breaks down alpha-D-galactose residues at the non-reducing ends of oligosaccharides (such as raffinose, stachyose, and verbascose), complex galactomannans, and galactolipids.

Beta-mannans are insoluble plant polysaccharides with beta-1,4-linked mannose as the backbone. We used three forms of this polysaccharide, namely, pure mannan, glucomannan, and galactomannan, to enrich haloarchaea, which have the ability to utilize mannans for growth. Four mannan-utilizing strains obtained in pure cultures were closely related to each other on the level of the same species.
