A Cu,Zn superoxide dismutase (SOD1) gene was cloned from the bumblebee, Bombus ignitus. The SOD1 gene of B. ignitus spans 1,317 bp and consists of three introns and four exons encoding 151 amino acid residues. The B. ignitus SOD1 (BiSOD1) possesses the typical metal-binding ligands of six histidines and one aspartic acid common to SOD1s. The deduced amino acid sequence of the BiSOD1 cDNA showed 82% identity to Apis mellifera SOD1 and 68-64% to SOD1 sequences from other insects. Northern blot analysis revealed the presence of BiSOD1 transcripts in all tissues examined. When paraquat (methyl viologen), a free radical-inducing agent, was injected into the body cavity of B. ignitus workers, BiSOD1 mRNA expression was up-regulated in the fat body. In addition, the expression levels of BiSOD1 mRNA in the fat body significantly increased when B. ignitus workers were exposed to low (4 degrees C) or high (37 degrees C) temperatures, or injected with lipopolysaccharide (LPS), which suggests that BiSOD1 possibly protects against oxidative stress caused by extreme temperatures and bacterial infection.

The dopaminergic system is involved in caste-specific behaviors in eusocial bumble bees. However, little is known about how the caste differences in dopaminergic system are formed during pupal stages in the brains of bumble bees. Thus, we investigated the levels of dopamine-related substances and expression of genes encoding enzymes involved in dopamine synthesis and metabolism, dopamine receptors, and a dopamine transporter in the brain of female Bombus ignitus.

Bees have been known to visit the male-fertile cultivars of self-incompatible flowering plants more frequently than the male-sterile cultivars, but the origin of this preference is poorly understood. Here, we demonstrate that this preference is driven by the higher protein/lipid ratio of male-fertile pollen compared with male-sterile pollen by way of two caged-behavioral assays with six cultivars. In the first assay, flower-naïve bumblebees (Bombus ignitus Smith) showed a significantly higher flower-visitation rate to male-fertile cultivars (pollen germination rate > 55%; > 14 visits/10 min) of the Japanese pear (Pyrus pyrifolia Nakai) than male-sterile cultivars (pollen germination rate ≤ 20%; > 6 visits/10 min).

Since the publication of Sprengel's (1793) observations, it has been considered that flowers with zygomorphic (or bilaterally symmetrical) corollas evolved to restrict the movement of pollinators into the flower by limiting the pollinator's direction of approach. However, little empirical support has been accumulated so far. Our aim was to build on previous research that showed zygomorphy reduces variance in pollinator entry angle, aiming to observe whether floral symmetry or orientation had an impact on pollinator entry angle in a laboratory experiment using bumble bees, Bombus ignitus.

Four insect orders have flight muscles that are both asynchronous and indirect; they are asynchronous in that the wingbeat frequency is decoupled from the frequency of nervous stimulation and indirect in that the muscles attach to the thoracic exoskeleton instead of directly to the wing. Flight muscle thick filaments from two orders, Hemiptera and Diptera, have been imaged at a subnanometer resolution, both of which revealed a myosin tail arrangement referred to as “curved molecular crystalline layers”. Here, we report a thick filament structure from the indirect flight muscles of a third insect order, Hymenoptera, the Asian bumble bee Bombus ignitus.

To compare the behavioral roles of biogenic amines in the males of primitive and advanced eusocial bees, we determined the levels of dopamine- and octopamine-related substances in the brain, and the behavioral effects of these monoamines by drug injection in the primitive eusocial bumble bee, Bombus ignitus. The levels of dopamine and its precursors in the brain peaked at the late pupal stage, but the dopamine peak extended to adult emergence. The tyramine and octopamine levels increased from the mid-pupal to adult stages.
