Kinetics and the mechanism of total in vitro hydrolyses (i.e. up to the exhaustion of substrate) of acetylcholine and acetylthiocholine by acetylcholinesterase and butyrylcholinesterase were studied in vitro in a batch reactor at 25 degrees C, pH 8 and ionic strength of 0.11 M. Every hydrolysis was monitored by 2-3 independent analytical methods. All studied types of enzymatic hydrolyses fulfilled the Michaelis-Menten reaction scheme with the irreversible second step. A table of obtained average values of rate constants and estimations of initial molar enzyme concentrations, and discussion of the results are presented.

It is of great significance for the clinical diagnosis of Alzheimer's disease (AD) to achieve the on-site activity evaluation of acetylcholinesterase (AChE), the hydrolase of acetylcholine (ACh). Herein, we have developed a biosensing method endowed with considerable superiority based on the organic-inorganic hybrid composite Eu(DPA)@Lap with excellent stability and fluorescent properties for this purpose by loading Eu ions and 2,6-dipicolinic acid (DPA) into LAPONITE® (Lap). Through the comprehensive consideration of the specific hydrolysis of acetylthiocholine (ATCh) into thiocholine (TCh) by AChE, the high binding affinity of TCh to copper ion (Cu), and the selective fluorescence quenching ability of Cu, a simple Eu(DPA)@Lap-based assay was developed to realize the rapid and convenient evaluation of AChE activity.

A novel colorimetric biosensor for the sensitive and selective detection of an organophosphate pesticide, paraoxon ethyl (POE), was developed based on its inhibitory effect on the acetylcholine esterase (AChE) enzyme. The bis-neocuproine copper (II) complex ([Cu(Nc)]) known as the CUPRAC reagent, was used as a chromogenic oxidant in the AChE inhibition-based biosensors for the first time.
