Background: It is unclear whether a stimulus that cannot be recognized consciously, could elicit a well-processed cognitive response.
Methods: We used functional imaging to examine the pattern of cortical activation elicited by unrecognized stimuli during memory processing. Subjects were given a recognition task using recognizable and non-recognizable subliminal stimuli.
Results: Unrecognized stimuli activated the cortical areas that are associated with retrieval attempt (left prefrontal), and novelty detection (left hippocampus). This indicates that the stimuli that were not consciously recognized, activated neural network associated with aspects of explicit memory processing.
Conclusion: Results suggest that conscious recognition of stimuli is not necessary for activation of cognitive processing.

Lysosomes are best known for their roles in inflammatory responses by engaging in autophagy to remove inflammasomes. Here, we describe an unrecognized role for the lysosome, showing that it finely controls macrophage inflammatory function by manipulating the lysosomal Fe-prolyl hydroxylase domain enzymes (PHDs)-NF-κB-interleukin 1 beta (IL1B) transcription pathway that directly links lysosomes with inflammatory responses. TRPML1, a lysosomal cationic channel, is activated secondarily to ROS elevation upon inflammatory stimuli, which in turn suppresses IL1B transcription, thus limiting the excessive production of IL-1β in macrophages.

Rapid neutrophil recruitment is critical for controlling infection, with dysfunctional neutrophil responses in diseases like diabetes associated with greater morbidity and mortality. We have shown that the leukocyte protein ECRG4 enhances early neutrophil recruitment to cutaneous wounds and hypothesized that ECRG4 regulates the early host response to infection. Using a cutaneous infection model, we found that ECRG4 KO mice had decreased early neutrophil recruitment with persistent larger lesions, increased bacterial proliferation and systemic dissemination.
