The adenylate cyclase (cyaA) gene of Bordetella pertussis is not expressed in Escherichia coli. Using cya-lac fusions, high-expression spontaneous mutants were isolated and shown to have the insertion element IS2 in orientation II integrated into the reading frame of cyaA. Upon transfer of the IS2-activated cya-lac fusion into B. pertussis, we found that the IS2-provided promoter is as efficient in B. pertussis as it is in E. coli. These results provide evidence that an insertion element derived from the E. coli chromosome can activate gene expression in B. pertussis, a taxonomically distant organism.

Noncoding satellite DNA repeats are abundant at the pericentromeric heterochromatin of eukaryotic chromosomes. During interphase, sequence-specific DNA-binding proteins cluster these repeats from multiple chromosomes into nuclear foci known as chromocenters. Transposable elements are DNA sequences that can move and replicate within genomes. Broadly, there are 2 types: autonomous elements, which encode the necessary enzymes for transposition, and nonautonomous elements, which rely on the enzymes produced by autonomous elements for their transposition. Nonautonomous elements have been proposed to regulate the numbers of transposable elements, which is a possible explanation for the persistence of transposition activity over long evolutionary times.

In order to systematically study the bending behavior of the connection joints of the disc-buckle type formwork support, the accurate numerical model of the disc-buckle type connection joints was obtained through the experimental on the bending behavior of the connection joints of the disc-buckle type, and the bending moment-rotation curve of the joints was verified. The analysis of the failure mode and stress distribution of the joints reveals the importance of the bending behavior of each component. By establishing an accurate numerical model of the joint, the accuracy of the bending experiment of the joint was verified, and the parametric analysis of the influence factors such as the depth of the wedge insertion the disk-plate, the initial position of the wedge insertion the disk-plate, the thickness of the wedge, material constitutive of the wedge and the thickness of the disk-plate was carried out to grasp the influence of the relevant parameters on the bending behavior of the joint.
