Proline plays a central role in the biosynthesis of prodigiosin by Serratia marcescens. Proline catabolism takes place by oxidation catalysed by the enzyme proline oxidase encoded by the gene putA. A gene bank of chromosomal DNA from S. marcescens was constructed using the plasmid vector pBR328, and then recombinant DNA was used in transformation experiments with Escherichia coli HB 101 as recipient strain. One of the recombinant plasmids, pSL001, was encoded for proline oxidase. Subcloning experiments led to a second plasmid pSL008 able to maintain proline oxidase activity.

Novel and simple spectrophotometric and distance based procedures for thiols (L-cysteine, N-acetylcysteine, and glutathione) determination in biological fluids and pharmaceuticals have been proposed based on their inhibitory action on the oxidation of catechol in the presence of Agaricus bisporus crude extract (ABE). The influence of L-glycine, L-alanine, L-proline, L-methionine, L-cystine, ascorbic acid, uric acid, and bilirubin on the thiol determination has been investigated. Uric acid, bilirubin, L-cystine (oxidized thiol), and L-amino acids do not interfere with the determination.

In bacteria the formation of disulphide bonds is facilitated by a family of enzymes known as the disulphide bond forming (Dsb) proteins, which, despite low sequence homology, belong to the thioredoxin (TRX) superfamily. Among these enzymes is the disulphide bond-forming protein A (DsbA); a periplasmic thiol oxidase responsible for catalysing the oxidative folding of numerous cell envelope and secreted proteins. Pathogenic bacteria often contain diverse Dsb proteins with distinct functionalities commonly associated with pathogenesis.
