Calcium binding by gadolinium-based MR contrast agents.

Purpose: This study was conducted to characterize the alterations in ionic sodium, potassium, and calcium by gadolinium-based MR contrast agents.

Materials And Methods: An electrolyte solution (ES) containing 1.2 mM/L calcium ions,120 mM/L sodium, and 4.0 mM/L potassium were diluted with various gadolinium compounds and alterations in ionized electrolytes were measured using an ion-specific electrometer. Gadolinium compounds including Gd-DTPA, Gd-DOTA, gadoteridol, gadodiamide, meglumine/sodium diatrizoate (76% Urografin), and isotonic saline as a control were investigated. The dilution ranged from 5% (ES/test solution = 100/5) to 100%. Alterations of ionic electrolytes were measured. Calcium-binding capacities caused by each gadolinium compound also were measured.

Results: The alterations of ionic sodium and potassium by gadolinium compound were similar to those of isotonic saline. A significant reduction in ionized calcium was observed with Gd-DTPA and Gd-DOTA in comparison with gadoteridol and gadodiamide.

Conclusion: Ionic gadolinium compounds induced significant reductions of calcium ions in vitro compared with non-ionic gadolinium compounds.

This study introduces a series of water-soluble radical dendrimers (G0 to G5) as promising magnetic resonance imaging (MRI) contrast agents that could potentially address clinical safety concerns associated with current gadolinium-based contrast agents. By using a simplified synthetic approach based on a cyclotriphosphazene core and lysine-derived branching units, we successfully developed a G5 dendrimer containing up to 192 units of 2,2,6,6-Tetramethylpiperidinyloxy (TEMPO) radical. This synthesis offers advantages including ease of preparation, purification, and tunable water solubility through the incorporation of glutamic acid anion residues.

This proof-of-concept study explores quantitative imaging of articular cartilage using photon-counting detector computed tomography (PCD-CT) with a dual-contrast agent approach, comparing it to clinical dual-energy CT (DECT). The diffusion of cationic iodinated CA4 + and non-ionic gadolinium-based gadoteridol contrast agents into ex vivo bovine medial tibial plateau cartilage was tracked over 72 h. Continuous maps of the contrast agents' diffusion were created, and correlations with biomechanical indentation parameters (equilibrium and instantaneous moduli, and relaxation time constants) were examined at 28 specific locations.

The use of hepatobiliary-specific contrast agents in liver MRI is a crucial diagnostic tool for evaluating liver disease, enabling the detection and characterisation of focal lesions and vascular alterations, as well as the assessment and grading of chronic hepatopathy. Paramagnetic hepatobiliary-specific contrast agents are gadolinium-based, partially taken up by hepatocytes, and excreted via both renal and biliary pathways. There are two linear ionic molecules that are currently commercially available: gadobenic acid (Gd-BOPTA) and gadoxetic acid (Gd-EOB-DTPA).

The incidence of post-contrast acute kidney injury (PC-AKI) is low with the new low- or iso-osmolar non-ionic iodinated contrast agents. The only proven form of prophylaxis for PC-AKI is hydration, preferably intravenous, although oral hydration is equally effective. In this article we define PC-AKI and its risk factors, propose a prophylaxis protocol and respond to the most common doubts that arise around prophylaxis.
