We describe a simple, sensitive and noninvasive assay that uses nontoxic, reengineered anthrax toxin-beta-lactamase fusion proteins with altered protease cleavage specificity to visualize specific cell-surface proteolytic activity in single living cells. The assay could be used to specifically image endogenous cell-surface furin, urokinase plasminogen activator and metalloprotease activity. We have adapted the assay for fluorescence microscopy, flow cytometry and fluorescent plate reader formats, and it is amenable for automation and high-throughput analysis.

CyHV-2 is the pathogen of herpesvirus haematopoietic necrosis (HVHN), resulting in significant economic losses in crucian carp. Although multiple oral vaccines have been developed to prevent CyHV-2, they have not achieved ideal protective effects. To improve the protective effect of oral vaccine, a combination vaccine was conducted by mixing recombinant Saccharomyces cerevisiae displaying ORF132 or ORF25 on the cell surface in a 1:1 ratio.

Many brain-targeting drug delivery strategies have been reported to permeate the blood-brain barrier (BBB) via hijacking receptor-mediated transport. However, these receptor-based strategies could mediate whole-brain BBB crossing due to the wide intracranial expression of target receptors and lead to unwanted accumulation and side effects on healthy brain tissues. Inspired by brain metastatic processes and the selectivity of brain metastatic cancer cells for the inflammatory BBB, a biomimetic nanoparticle was developed by coating drug-loaded core with the inflammatory BBB-seeking erythrocyte-brain metastatic hybrid membrane, which can resist homotypic aggregation and specially bind and permeate the inflammatory BBB for specific drug delivery.

Phosphatase and Tensin Homologue (PTEN) is one of the most frequently lost tumor suppressors in cancer and the predominant negative regulator of the PI3K/AKT signaling axis. A growing body of evidence has highlighted the loss of PTEN with immuno-modulatory functions including the upregulation of the programmed death ligand-1 (PD-L1), an altered tumor derived secretome that drives an immunosuppressive tumor immune microenvironment (TIME), and resistance to certain immunotherapies. Given their roles in immunosuppression and tumor growth, we examined whether the loss of PTEN would impact the biogenesis, cargo, and function of extracellular vesicles (EVs) in the context of the anti-tumor associated cytokine interferon-γ (IFN-γ).

This study aimed to examine the extraction of specific phenolic compounds from blackthorn using ultrasound-assisted extraction (UAE) and to evaluate the influence of UAE on the phenolic composition, bioaccessibility, and cytotoxic effect evaluated on ovarian cancer (OVCAR-3 and SKOV-3) and healthy (HaCaT) cell lines. The UAE parameters were optimized by modeling with the response surface method. Temperature, time, and ultrasound amplitude were utilized to determine the optimal extraction conditions.
