The incorporation of butyl-methoxydibenzoylmethane (BMDBM), one of the most efficient and frequently used UV-A blockers, into lipospheres was examined in order to decrease the light-induced sunscreen degradation. Lipospheres, obtained by the melt technique and using tristearin as the lipid material and hydrogenated phosphatidylcholine as the emulsifier, showed proper features in terms of size (10-40 microm), BMDBM loading level (21.63% +/- 0.90%, w/w) and physical state. Photolysis studies, involving irradiation of lipospheres with simulated sunlight before and after their introduction in emulsion formulations, demonstrated a relevant enhancement of the encapsulated sunscreen photostability in comparison with unencapsulated BMDBM.

The incorporation of drug substances into the matrix of solid lipid microparticles (SLM) is critical to providing effects such as prolonged release, taste masking, and protection of the labile API. Currently, a commonly used method of characterizing multi-compartment lipid systems, such as SLM, is to determine entrapment efficiency (EE) and drug loading (DL) parameters, but this is not sufficient for understanding the localization of API either in the core or on the surface of the microspheres. The main objective of the research was to study the distribution of API in an aqueous dispersion of SLM in order to distinguish between the API incorporated in the lipid matrix and localized in the superficial region (interphase) and to refer the obtained results to the EE and DL parameters.

Fenofibrate lipospheres were prepared by the melt dispersion technique. Critical parameters influencing particle size and entrapment efficiency were optimized by applying the L9 Taguchi experimental design. Entrapment efficiency of up to 87 % was obtained for the optimized formulation on increasing olive oil up to 30 % in the lipid carrier.

Objective: Preparation and characterization of an oral pro-dispersion liposphere formulation for cyclosporin, a water insoluble drug with limited bioavailability.

Methods: Pro-dispersion formulation consisted of a solid fat, dispersing agents and amphiphilic solvents as the major components besides cyclosporin A (CsA) were prepared in the present work. For preparation of this formulation, phospholipid was dissolved in pharmaceutically acceptable water soluble organic solvent, thereafter CsA along with other components was added and formulation optimization was carried out.
