Photo-CIDNP effects interpreted for individual residues are used for the structural characterization of non-native ensembles of proteins, which is described in this paper. Two-dimensional photo-CIDNP experiments are compared to conventional HSQC spectra to elucidate the relative solvent exposure of the six tryptophan residues in non-native states of hen egg white lysozyme. The differential solvent accessibility of the tryptophan residues in non-native lysozyme coincides with the dynamical properties of these residues monitored for both backbone and side chain NH sides obtained from analysis of transverse relaxation measurements. These data can be interpreted in the context of the hydrophobic clustering around the tryptophan residues and is supported by the application of this method to the cluster breaking W62G mutant of lysozyme.

Hence, the chemoselective functionalization of tryptophan residue can provide access to site-selective bioconjugation of almost all known proteins. With the increase in the utility of bioconjugated proteins and peptides as drugs and therapeutic agents, the development of smart protocols to fabricate and modulate biomolecules has flourished.

The structural groups of 2-oxindole and tricyclic 3a-hydroxy-hexahydropyrrolo-[2,3-]indole (HO-HPI) are important pharmacophores. Chemical synthesis of complex alkaloids containing a 2-oxindole or HO-HPI moiety, especially the latter one, has been a long-standing challenge. Herein, we characterized the P450 enzyme AfnD, and its homologue proteins, HmtT, ClpD, KtzM, and LtzR, as cyclopeptide 2-oxindole and HO-HPI monooxygenases (cpOPMOs) that could introduce a 2-oxindole or HO-HPI moiety into the tryptophan-containing cyclopeptides in a pH-dependent manner.
