Published: April 2006

A series of novel benzoxazole benzenesulfonamides was synthesized as inhibitors of fructose-1,6-bisphosphatase (FBPase-1). Extensive SAR studies led to a potent inhibitor, 53, with an IC(50) of 0.57microM. Compound 17 exhibited excellent bioavailability and a good pharmacokinetic profile in rats.

Article Synopsis
  • * The study revealed that this exposure significantly reduced the areas under the curves (AUC) of probe substrates for cytochrome P450 enzymes, indicating decreased drug absorption and effectiveness.
  • * Mechanisms for reduced drug absorption were linked to decreased intestinal permeability and impaired gut motility, suggesting potential drug-drug interactions that could lower the effectiveness of coadministered medications.

Sulfonamide compounds, also known as sulfa drugs, are a significant class of synthetic bacteriostatic antimicrobials and were the primary source of therapy against bacterial infections before the introduction of penicillin in 1941. Hybridization of sulfonamides with various pharmaceutically active heterocyclic moieties leads to sulfonamide hybrids with a wide variety of biological activities. Part B of this review presents the most recent advances in designing and developing more two-component sulfonamide hybrids containing triazole, thiadiazole, triazine, oxazole/ benzoxazole, isoxazole, oxadiazole, imidazole, benzimidazole, furan, benzofuran, thiophene, pyrrole, indazole, tetrazole, chromene/ chromone, pyridazine, quinoxaline, acridine, phthalazine, and xanthone between 2015 and 2020.

Introduction: Pharmacokinetic variability in disease state is common in clinical practice, but its underlying mechanism remains unclear. Recently, gut microbiota has been considered to be pharmacokinetically equivalent to the host liver. Although some studies have explored the roles of gut microbiota and host Cyp450s in drug pharmacokinetics, few have explored their effects on pharmacokinetic variability, especially in disease states.

Hemoglobin-vesicles (Hb-V) are being developed as red blood cell (RBC) substitutes. In this study, we report on quantitative and qualitative alterations of hepatic cytochrome P450 (CYPs) and the pharmacokinetics of CYP-metabolizing drugs, with a focus on four CYP isoforms (CYP1A2, CYP2C11, CYP2E1 and CYP3A2), after Hb-V resuscitation from a massive hemorrhage. The results of proteome analysis and western blot data indicate that resuscitation with both Hb-V and packed RBC (PRBC) resulted in a decrease in the protein levels of CYPs.
