B cell development in the bone marrow is a highly regulated process and expression of a functional pre-BCR represents a crucial checkpoint, common to human and mouse. In this review, we discuss pre-BCR analogies and differences between the two species leading to pre-B cell differentiation and proliferation. In addition, the mechanisms triggering pre-BCR activation are reviewed, taking into account the recent report of heparan sulfates and galectin 1 as stromal cell-derived pre-BCR ligands. Finally, ligand-induced pre-BCR activation models are proposed on the bases of the differences reported for pre-BCR and IL7 dependencies in the two species.

Article Synopsis
  • The study investigates the role of ERBB3 expression in HER2-positive breast cancer and its potential impact on treatment resistance.
  • Through various bioinformatics techniques and machine learning algorithms, the researchers identified three key genes—PBX1, IGHM, and CXCL13—that are linked to ERBB3 expression and can serve as prognostic markers.
  • Findings suggest that these genes may influence breast cancer cell behavior and prognosis, emphasizing their significance in understanding HER2-positive breast cancer treatment outcomes.

STAT1 Promotes PD-L1 Activation and Tumor Growth in Lymphangioleiomyomatosis.

Lymphangioleiomyomatosis (LAM) is a cystic lung disease that primarily affects women. LAM is caused by the invasion of metastatic smooth muscle-like cells into the lung parenchyma, leading to abnormal cell proliferation, lung remodeling and progressive respiratory failure. LAM cells have TSC gene mutations, which occur sporadically or in people with Tuberous Sclerosis Complex.

Article Synopsis
  • - Eosinophils are key players in asthma inflammation, releasing harmful proteins like major basic protein (MBP) that damage airway cells and contribute to asthma symptoms.
  • - Poly-l-arginine (PLA), which mimics MBP, induces a new cell death mechanism called ferroptosis in airway cells by reducing GABARAPL1 levels, which is linked to eosinophil activity in asthma.
  • - Targeting the relationship between eosinophils, MBP, and the mTORC1/PBX1/GABARAPL1 signaling pathway could provide new ways to treat asthma by reducing inflammation and preventing cell death in the airway epithelium.

How to think about acute leukemia of ambiguous lineage.

Hematology Am Soc Hematol Educ Program

December 2024

UT Southwestern Medical Center, BioCenter, Dallas, TX.

Classification of acute leukemia involves assigning lineage by resemblance of blasts to normal progenitor cells. This approach provides descriptive information that is useful for disease monitoring, provides clues to pathogenesis, and can help to select effective chemotherapeutic regimens. Acute leukemias of ambiguous lineage (ALAL) are those leukemias that either fail to show evidence of myeloid, B-lymphoid, or T-lymphoid lineage commitment or show evidence of commitment to more than 1 lineage, including mixed-phenotype acute leukemia (MPAL).
