The activation of Hedgehog (Hh) signaling has been implicated in the growth of various tumor types, including gastric carcinoma. However, the precise mechanisms of Hh activation and suppression of tumor growth by the blockade of Hh signaling in gastric carcinoma cells remain unknown. The aim of this study was to elucidate the mechanism of abnormal Hh signaling and the key molecules contributing to dysregulated growth of gastric carcinoma. The Sonic hedgehog (Shh) ligand and its receptor Patched were expressed in all five gastric carcinoma cell lines examined (MKN1, MKN7, MKN45, MKN74, and AGS cells). The blockade of Hh signaling with anti-Shh antibody inhibited the growth of all five gastric carcinoma cell lines. Shh was overexpressed (mean, 12.8-fold) in 8 of 14 (57.0%) cancerous tissue samples from patients with gastric carcinoma as compared with expression in the surrounding noncancerous tissues. The disruption of glioma-associated oncogene 1 (Gli1) by small interfering RNA induced an increase in p21/cyclin-dependent kinase-interacting protein 1 (CIP1), interfered with the G1-S transition, and suppressed cell proliferation. The stimulation or inhibition of Hh signaling did not affect p53 activity and the induction of p21/CIP1 expression and the G1 arrest by inhibition of Hh signaling were not affected by the p53 status. These findings suggest that the overexpression of Shh contributes to constitutive Hh activation and that this signaling pathway negatively regulates p21/CIP1 through a Gli1-dependent and p53-independent mechanism in gastric carcinoma cells.

Trousseau's syndrome is a thromboembolic disorder associated with malignancies, with cerebral infarction and hemorrhage representing common central nervous system complications in patients with cancer. This report details the diagnosis and treatment of a patient with gastric adenocarcinoma at our institution who concurrently developed cerebral infarction and subarachnoid hemorrhage. We performed a comprehensive literature review in the Wanfang and PubMed databases, searching for relevant studies on Trousseau's syndrome, cerebral embolism, and subarachnoid hemorrhage.

PLASMA: Partial LeAst Squares for Multiomics Analysis.

Cancers (Basel)

January 2025

Department of Biostatistics, Data Science, and Epidemiology, School of Public Health, Georgia Cancer Center at Augusta University, Augusta, GA 30912, USA.

: Recent growth in the number and applications of high-throughput "omics" technologies has created a need for better methods to integrate multiomics data. Much progress has been made in developing unsupervised methods, but supervised methods have lagged behind. : Here we present the first algorithm, PLASMA, that can learn to predict time-to-event outcomes from multiomics data sets, even when some samples have only been assayed on a subset of the omics data sets.

Harnessing Machine Learning and Nomogram Models to Aid in Predicting Progression-Free Survival for Gastric Cancer Patients Post-Gastrectomy with Deficient Mismatch Repair(dMMR).

BMC Cancer

Department of Gastroenterology, Shanxi Hospital Affiliated to Carcinoma Hospital, Chinese Academy of Medical SciencesShanxi Province Carcinoma Hospital, Carcinoma Hospital Affiliated to Shanxi Medical University, Taiyuan, Shanxi, 030013, People's Republic of China.

Objective: To assess the effectiveness of a machine learning framework and nomogram in predicting progression-free survival (PFS) post-radical gastrectomy in patients with dMMR.

Method: Machine learning models and nomograms to forecast PFS in patients undergoing radical gastrectomy for nonmetastatic gastric cancer with dMMR. Independent risk factors were identified using Cox regression analysis to develop the nomogram.

Incidence and Heritability of Gastric Carcinoma in the Belgian Shepherd Dog Population in The Netherlands.

Vet Sci

January 2025

Department of Clinical Sciences of Companion animals, Faculty of Veterinary Medicine, Utrecht University, 3584 CL Utrecht, The Netherlands.

Background: Gastric carcinoma in the Tervueren and Groenendael varieties of the Belgian Shepherd dog is commonly recognized and carries a grave prognosis. Information on incidence and heritability is necessary to design an effective selection strategy against this disease.

Methods: A database of cases and controls was built including cases with Tier 1 (confirmed via endoscopy or post-mortem) and Tier 2 evidence of gastric carcinoma.
