GASepo-a software solution for quantitative analysis of digital images in Epo doping control.

A software has been developed that is aimed at quantitative analysis of images acquired by isoelectric focusing and double blotting procedures used for recombinant erythropoietin doping control. It represents a unified and easy-to-use tool for Epo doping experts in WADA accredited laboratories. It is based on image segmentation philosophy that enables identification of individual bands whose characteristics are needed for evaluation of the Epo doping positivity criteria. Several modules implemented in the GASepo software include an original know how, in particular, the method of robust calculation of the cut-off line, band segmentation and classification algorithms. GASepo is being used in several doping control laboratories worldwide.

In this study, the performance of the new MAIIA EPO purification kit 7D3 was evaluated for human urine samples to confirm compliance with the WADA criteria of the WADA technical document 2024 (TD2024EPO). The kit was validated for selectivity, reliability at minimum required performance levels (MRPLs), limit of detection (LOD), carryover and recovery. A total of 81 urine samples were tested for selectivity, 29 samples for reliability at MRPL, LOD and 16 samples for recovery.

Detection and monitoring of biomarkers related to doping is particularly suitable for the development of analytical strategies dedicated to indirect detection of banned substances. Previous studies in horses have already allowed the investigation of transcriptomic biomarkers in equine blood associated with reGH and rHuEPO administrations. Our most recent developments continue to focus on the discovery and monitoring of transcriptomic biomarkers for the control of ESAs, and a collaborative study with WADA-accredited doping control laboratories has recently been initiated to conduct a pilot study.
