A new approach to oligonucleotide N3'-->P5' phosphoramidate building blocks.

Nucleosides Nucleotides Nucleic Acids

Geron Corporation, Menlo Park, California, USA.

Published: December 2005

A new synthetic approach to 5-phosphoramidites of 3'-aminonucleosides was developed. The methodology relies upon the use of 3'-amino-2',3'-dideoxy nucleosides as the key starting materials. The final phosphoramidite products were obtained with high yields via 2-3-step efficient chemical transformations using selective introduction of orthogonal protective groups to the 3'-aminonucleoside sugar and base moieties.

Gene therapy in polycystic kidney disease: A promising future.

J Transl Int Med

Polycystic kidney disease (PKD) is a genetic disorder marked by numerous cysts in the kidneys, progressively impairing renal function. It is classified into autosomal dominant polycystic kidney disease (ADPKD) and autosomal recessive polycystic kidney disease (ARPKD), with ADPKD being more common. Current treatments mainly focus on symptom relief and slowing disease progression, without offering a cure.

Nucleophilic Reactions of Phosphorothioate Oligonucleotides.

Small Methods

January 2025

College of Pharmaceutical Sciences, Southwest University, Chongqing, 400715, China.

The nucleophilic reaction between phosphorothioate oligonucleotides and electrophilic reagents has become a cost-effective and efficient approach for oligonucleotide functionalization. This method allows for the precise incorporation of desired chemical structures at specific sites on the phosphorothioate backbone through conjugation with electrophilic groups. The reaction is characterized by its high reactivity and yield, as well as its ability to enhance the hydrophilicity of otherwise hydrophobic compounds.

Article Synopsis
  • There's an urgent need to boost the effectiveness of seasonal influenza vaccines, with recombinant hemagglutinin showing potential benefits over traditional methods.
  • In a study, two adjuvants (Advax-CpG55.2 and alum-CpG55.2) were tested to see if they could improve the immune response of a quadrivalent influenza vaccine (QIV) in mice.
  • Results showed that the adjuvanted vaccines led to much higher levels of protective antibodies and significantly reduced illness in mice infected with the H1N1 strain, suggesting that this approach could be a better alternative for seasonal flu vaccinations.

Directly detecting biomarkers in liquid biopsy for diagnosis and personalized treatment plays a crucial role in managing cancer relapse and increasing survival rates. Typically, the standard analysis of circulating tumour DNA requires lengthy isolation, extraction, and amplification steps, leading to sample contamination, longer turnaround time and higher assay costs. Surface plasmon resonance is an emerging and promising technology for rapid and real-time dynamic biomarker monitoring in liquid biopsy.

In this article, we present an approach to maximizing the splicing regulatory properties of splice-switching oligonucleotide (SSO) designed to regulate alternative splicing of PKM pre-mRNA. The studied SSO interacts with the regulatory element in exon 10 of PKM pre-mRNA and contributes to a significant reduction of PKM2 level with a simultaneous increase of the PKM1 isoform. This SSO forms a duplex not only with the regulatory fragment of exon 10 but also with a similar RNA fragment of intron 9.
