Characterization of the Family I inorganic pyrophosphatase from Pyrococcus horikoshii OT3.

A gene encoding for a putative Family I inorganic pyrophosphatase (PPase, EC 3.6.1.1) from the hyperthermophilic archaeon Pyrococcus horikoshii OT3 was cloned and the biochemical characteristics of the resulting recombinant protein were examined. The gene (Accession No. 1907) from P. horikoshii showed some identity with other Family I inorganic pyrophosphatases from archaea. The recombinant PPase from P. horikoshii (PhPPase) has a molecular mass of 24.5 kDa, determined by SDS-PAGE. This enzyme specifically catalyzed the hydrolysis of pyrophosphate and was sensitive to NaF. The optimum temperature and pH for PPase activity were 70 degrees C and 7.5, respectively. The half-life of heat inactivation was about 50 min at 105 degrees C. The heat stability of PhPPase was enhanced in the presence of Mg2+. A divalent cation was absolutely required for enzyme activity, Mg2+ being most effective; Zn2+, Co2+ and Mn2+ efficiently supported hydrolytic activity in a narrow range of concentrations (0.05-0.5 mM). The K(m) for pyrophosphate and Mg2+ were 113 and 303 microM, respectively; and maximum velocity, V(max), was estimated at 930 U mg(-1).

Organic-inorganic hybrid ferroelectric compounds of the halobismuthate family have emerged as a focal point of research owing to their reduced toxicity and distinctive optical characteristics. This study presents a novel ammonium hybrid perovskite, [BPMBDMA]·[Bi2Br9], which exhibits both ferro- and piezoelectric properties and crystallizes in the polar noncentrosymmetric 2 space group. The nonlinear optical (NLO) activity of [BPMBDMA]·[Bi2Br9] was corroborated through second harmonic generation measurements evidencing its noncentrosymmetric structure, which was further substantiated by piezoresponse force microscopy analyses.

Lanthanide-based Single-Molecule Magnets (SMMs) with optical and magnetic properties provide a means to understand intrinsic energy levels of 4f ions and their influence on optical and magnetic behaviour. Hybrid coordination polimers based on AgX (with X = Cl, Br) and 2-, 3-, 4-picolylamine ligands, obtained by means of solvent-free methods, show peculiar luminescence properties that are strongly influenced by their structural motif, which in turn is defined by the adopted isomer of the ligand. A comprehensive study, combining photophysical methods and DFT calculations, allowed to rationalize the emissive behaviour of such hybrid coordination polymers in relation to their crystal structures and electronic properties. By means of luminescence measurements at variable temperatures, the nature of the emissive excited states and their deactivation dynamics was interpreted, revealing XMLCT transitions in the [(AgX)(2-pica)] compounds, a TADF behaviour in the case of 3-pica derivatives, and a dual emission at room temperature for the [(AgX)(4-pica)] family.

Correlating Microbial Dynamics with Key Metabolomic Profiles in Three Submerged Culture-Produced Vinegars.

Although vinegar is a product obtained by a well-known bioprocess from a technical point of view, the complex microbiota responsible for its production and their involvement in the organoleptic profiles are not clear yet. In this work, three acetification profiles in submerged culture using both synthetic and raw materials from Andalusia (Spain) were characterized by metagenomic (16S rRNA amplicon sequencing) and metabolomic tools (stir-bar sorptive extraction with thermo-desorption coupled to gas chromatography-mass spectrometry (SBSE-TD-GC-MS) and high-performance liquid chromatography (HPLC)). A total of 29 phyla, 208 families, and many more genera were identified, comprising bacteria and archaea as well as 75 metabolites, including minor volatile compounds, amino acids, biogenic amines, and other nitrogenous compounds.
