Signal transducers and activators of transcription (STAT) are a family of transcription factors that regulate a broad range of cellular processes, such as proliferation, differentiation, and survival, in a large variety of cell types. Because of their regulation of diverse cellular functions, their aberrant activation is frequently associated with disease development, particularly oncogenic diseases. Much evidence exists to suggest that STAT proteins play a significant role in cellular transformation. However, which STAT proteins and to what extent they cause transformation and subsequent disease progression are topics currently being investigated. In this review, we will report on the findings concerning the involvement of Stat6 in the development of lymphoma and leukemia. Mounting evidence, in both patients and mouse models, supports a model where Stat6 is not a mere bystander, but rather, plays an active role in promoting a transformed phenotype.

To date, there are no specific or effective therapeutic agents to control inflammation and alleviate cadmium-induced progressive destruction of brain cells. Fluoroquinolones (FQs), widely used antimicrobials with effective blood-brain barrier penetration, show promise in being repurposed as anti-inflammatory drugs.

Reactive oxygen species exacerbate nonalcoholic steatohepatitis (NASH) by oxidizing macromolecules; yet how they promote NASH remains poorly understood. Here, we show that peroxidase activity of global hepatic peroxiredoxin (PRDX) is significantly decreased in NASH, and palmitic acid (PA) binds to PRDX1 and inhibits its peroxidase activity. Using three genetic models, we demonstrate that hepatic PRDX1 protects against NASH in male mice.

Fatty liver hemorrhagic syndrome (FLHS) in laying hens is a nutritional and metabolic disease involving liver enlargement, hepatic steatosis, and hepatic hemorrhage as the primary symptoms. The syndrome is prone to occur during the peak laying period of laying hens, which has resulted in significant economic losses in the laying hen breeding industry; however, the specific pathogenesis of FLHS remains unclear. Our group and previous studies have shown that bile acid levels are significantly decreased during the development of fatty liver and that targeted activation of bile acid-related signaling pathways is beneficial for preventing and treating fatty liver.
