The activity of alaglyzin, a new drug based on a clathrate complex of allapinine (well-known antiarrhythmic) with glycyrrhizic acid (GA), was studied on the models of arrhythmia induced by itraperitoneal injections of calcium chloride (CaCl2) and adrenaline (epinephrine) in rats. Alaglyzin is less toxic than allapinine (LD50 70 and 6 mg/kg, respectively), and the effective dose of the parent compound in the complex form is also about 12 times lower than that in the reference drug. The experiments showed a dose dependent action of alaglyzin. Pretreatment of the experimental animals with alaglyzin in a dose of 0.25 mg/kg prevented from the arrhythmia development in both tests, while a dose of 0.125 mg/kg was effective only in the CaCl2, test.

Opioid dependence is defined by an aversive withdrawal syndrome upon drug cessation that can motivate continued drug-taking, development of opioid use disorder, and precipitate relapse. An understudied but common opioid withdrawal symptom is disrupted sleep, reported as both insomnia and daytime sleepiness. Despite the prevalence and severity of sleep disturbances during opioid withdrawal, there is a gap in our understanding of their interactions.

Enhancing protein O-GlcNAcylation by pharmacological inhibition of the enzyme O-GlcNAcase (OGA) has been considered as a strategy to decrease tau and amyloid-beta phosphorylation, aggregation, and pathology in Alzheimer's disease (AD). There is still more to be learned about the impact of enhancing global protein O-GlcNAcylation, which is important for understanding the potential of using OGA inhibition to treat neurodegenerative diseases. In this study, we investigated the acute effect of pharmacologically increasing O-GlcNAc levels, using the OGA inhibitor Thiamet G (TG), in normal mouse brains.

Objective: To investigate the effects of Astragalus polysaccharide (APS) on skeletal muscle structure and function in D-galactose (D-gal)-induced C57BL/6J mice.
Methods: Eighteen male C57BL/6J mice of specific pathogen-free (SPF) grade, aged 8 weeks, were selected and divided into three groups: a control group (0.9% saline gavage for 16 weeks), a D-gal group (subcutaneous injection of 200 mg/kg D-galactose in the upper neck region, once daily for 8 weeks), and a D-gal + APS group (subcutaneous injection of 200 mg/kg D-galactose, once daily for 8 weeks, with concurrent administration of 100 mg/kg APS by gavage for 8 weeks).
