The 70% aqueous methanolic extract of the Peruvian plant Oryctanthus sp. was found to contain a novel saccharide of a diene alpha,omega-diacid. Compound 1 was identified as an inhibitor of the VEGF receptor. The structure of this compound was established based on NMR studies. Compound 1 inhibited ligand binding to the VEGF receptor with an IC50 of 5.0 microM.

Purpose: This study evaluates the efficacy of intravitreal injections (IVI) of faricimab in patients with neovascular age-related macular degeneration (nAMD) and retinal pigment epithelium detachment (RPED) resistant to other anti-VEGF agents.
Material And Methods: The study included 61 patients (61 eyes) with nAMD previously treated with aflibercept and/or brolucizumab IVIs. Three groups were formed: group 1 received aflibercept IVI (32 eyes), group 2 received brolucizumab IVI (14 eyes), and group 3 received aflibercept followed by brolucizumab IVI (15 eyes).

The vascular endothelial growth factor receptor 2 (VEGFR2) and the hepatocyte growth factor receptor (C-Met) are critical receptors for signaling pathways controlling crucial cellular processes such as cell growth, angiogenesis and tissue regeneration. However, dysregulation of these proteins has been reported in different diseases, particularly cancer, where these proteins promote tumour growth, invasiveness, metastasis and resistance to conventional therapies. The identification of dual inhibitors targeting both VEGFR-2 and c-Met has emerged as a strategic therapeutic approach to overcome the limitations and resistance mechanisms associated with single-target therapies in clinical settings.
