A sulfur-substituted analogue of trigalacturonic acid (3) was synthesized. The synthesis features the application of 3-cyano-3-(tert-butyldimethylsilyl)oxypropylthioether (CSP) as a novel protective group for thiols. This analogue was designed with the expectation that it would be a stable analogous substrate for endo-polygalacturonase isolated from Stereum purpureum based on computer modeling experiments. Surface plasmon resonance experiments revealed that 3 forms a stable complex with the target enzyme.

Modular access to sulfur substituted analogues of isocytosine photoredox catalysis.

Chem Commun (Camb)

August 2024

Academy of Scientific and Innovative Research (AcSIR), Ghaziabad-201002, India.

A photoredox approach for synthesizing sulfur-substituted analogues of isocytosine coupling of modular phenyl propargyl chloride with thiourea has been reported. The resulting product with an amine group was found amenable to various late-stage modifications, providing access to a broad range of sulfur-containing isocytosine derivatives.

TDDFT study on a fluorescent probe for distinguishing analogous thiols based on smiles rearrangement.

The complete excited-state sensing mechanism of a fluorescent probe capable of distinguishing cysteine/homocysteine and glutathione from analogous biological thiols has been investigated. Using a TDDFT method, the nature of the fluorescence differences in the detection of thiols by the probe has been explained at the molecular level. Calculation results imply that the probe undergoes photoinduced electron transfer (PET) from the fluorophore to the nitrobenzooxadiazole (NBD)-based acceptor in the excited state.

The reaction of mercaptoacetic acid esters with pentachloro-2-nitro-1,3-butadiene provides the appropriate precursors for the synthesis of 2,3,4-trisubstituted benzo[]quinolines. These heterocycles are easily accessible via a single-step reaction with naphthalen-1-amine or anthracen-1-amine as the precursor. Due to the steric bulk and high electron density ring, the ring closure of benzo[]quinolines takes place exclusively.

Protein degradation is a promising strategy for drug development. Proteolysis-targeting chimeras (PROTACs) hijacking the E3 ligase cereblon (CRBN) exhibit enormous potential and universal degradation performance due to the small molecular weight of CRBN ligands. In this study, the CRBN-recruiting PROTACs were explored on the degradation of oncogenic fusion protein BCR-ABL, which drives the pathogenesis of chronic myeloid leukemia (CML).
