The Rep68 protein from Adeno-Associated Virus (AAV) is a multifunctional SF3 helicase that performs most of the DNA transactions necessary for the viral life cycle. During AAV DNA replication, Rep68 assembles at the origin of replication, catalyzing the DNA melting and nicking reactions during the hairpin rolling replication process to complete the second-strand synthesis of the AAV genome. We report the cryo-electron microscopy structures of Rep68 bound to the adeno-associated virus integration site 1 in different nucleotide-bound states.

Developmental and epileptic encephalopathies constitute a group of severe epilepsies, with seizure onset typically occurring in infancy or childhood, and diverse clinical manifestations, including neurodevelopmental deficits and multimorbidities. Many have genetic aetiologies, identified in up to 50% of individuals. Whilst classically considered paediatric disorders, most are compatible with survival into adulthood, but their adult phenotypes remain inadequately understood.

In 2024, CD19-CAR T cells are ubiquitous in rheumatology, with incredible therapeutic results in cases of severe and refractory disease. This major advance confirms the interest of the B lymphocyte as a therapeutic target, and also suggests the real possibility of a drug-free remission, at the price of minor and minimal side effects for the time being. However, the necessary perspective is still lacking for a therapy that remains out of reach because of its price.

Introduction: Dengue is one of the most widespread arboviruses in Latin America and is now affecting areas previously free of transmission. The COVID-19 pandemic and climatic variations appear to have affected the incidence of the disease, abundance of vectors and health programs related to dengue in some countries.
Objective: To analyze the epidemiology of dengue in Paltas, Ecuador (2016-2022), compare the periods before and during the COVID-19 pandemic, examine entomological reports and discuss the possible implications of the COVID-19 pandemic and climatic variations.

Chest imaging in children presents unique challenges due to varying requirements across age groups. For chest radiographs, achieving optimal images often involves careful positioning and immobilisation techniques. Antero-posterior projections are easier to obtain in younger children, while lateral decubitus radiographs are sometimes used when expiratory images are difficult to obtain and for free air exclusion.
