Propranolol inhibits the human ether-a-go-go-related gene potassium channels.

Propranolol is a noncardioselective beta-adrenergic antagonist that has been recently reported to prolong the QTc interval on the surface electrocardiogram in humans when overdosed [Farhangi, V., Sansone, R.A. (2003). QTc prolongation due to propranolol overdose. Int. J. Psychiatry Med. 33, 201-202.]. To examine the underlying mechanisms for these clinical findings, we studied the effects of propranolol on the human cardiac potassium channels encoded by the ether-a-go-go-related gene (ERG) using the whole cell voltage-clamp technique. We found that propranolol blocked hERG currents in a concentration-dependent manner with an IC50 of 9.9+/-1.3 microM which is relevant to the predicted plasma level of propranolol in this case report. The present study demonstrated that propranolol can inhibit hERG channels. The interaction between propranolol and hERG channels could lead to delayed cardiac repolarization and might be a molecular mechanism for the previously reported QTc prolongation when propranolol is overdosed.

Phosphatidic acid phosphatase, a conserved eukaryotic enzyme that catalyzes the Mg-dependent dephosphorylation of phosphatidic acid to produce diacylglycerol, has emerged as a vital regulator of lipid homeostasis. By controlling the balance of phosphatidic acid and diacylglycerol, the enzyme governs the use of the lipids for synthesis of the storage lipid triacylglycerol and the membrane phospholipids needed for cell growth. The mutational, biochemical, and cellular analyses of yeast phosphatidic acid phosphatase have provided insights into the structural determinants of enzyme function with the understanding of its regulation by phosphorylation and dephosphorylation.

An in situ self-cleaning covalent organic framework featuring arylbiguanide arms (Aryl-BIG-COF) was first developed to remove emerging organic pollutants such as propranolol (PRO) from water. The main breakthroughs addressed the scarcity of functional active sites, the impracticality of ex situ regeneration, and the rapid recombination of electronhole pairs in the application of COFs. Owing to the directional capture ability and electronic structure regulation of the arylbiguanide arms, the adsorption capacity and photocatalytic degradation rate of the newly synthesized COF increased by nearly four and seven times, respectively.
