14-deoxyandrographolide as a platelet activating factor antagonist in bovine neutrophils.

14-Deoxyandrographolide (14-DAP) is a labdane diterpene isolated from Andrographis paniculata with previously reported calcium channel blocking activity. Its potential platelet activating factor (PAF) antagonistic activity in bovine neutrophils was assessed. 14-DAP, in concentrations between 10-100 microM, reduced the extracellular acidification rate and the intracellular alkalinization in a dose-dependent manner. In addition, 14-DAP reduced PAF-induced calcium flux in the presence of extracellular calcium, and tyrosine phosphorylation of a 44 kDa protein corresponding to the MAPK(ERK1). However, 14-DAP reduced the 3H-PAF binding with a Ki of 7.8 x 10 (- 9)M, and a Hill slope of 0.63, suggesting that there is more than one binding site for 14-DAP. We concluded that 14-DAP is an effective antagonist of PAF-mediated processes in bovine neutrophils, probably by virtue of its calcium channel blocking property.

Due to the productive and economic consequences of Repeat Breeder (RB) syndrome, the objectives of this study were to determine the prevalence and risk factors for subclinical endometritis (SE) and oviductal occlusion (OO) in RB cows, and to make a therapeutic approach for these pathologies. In 99 RB cows, endometrial cytologies were performed to assess the presence of SE (>5 % polymorphonuclear neutrophils), and the oviductal patency was checked using the phenolsulfonphthalein test. Body condition score was evaluated, and data from each animal were obtained from on-farm software (parity, calving date, artificial insemination (AI) date, number of AI, and occurrence of postpartum diseases).

Virus replication is not required for oncolytic bovine herpesvirus-1 immunotherapy.

Oncolytic viruses are a promising approach for cancer treatment where viruses selectively target and kill cancer cells while also stimulating an immune response. Among viruses with this ability, bovine herpesvirus-1 (BoHV-1) has several advantages, including observations suggesting it may not require viral replication for its anti-cancer effects. We previously demonstrated that binding and penetration of enveloped virus particles are sufficient to trigger intrinsic and innate immune signaling in normal cells, while other groups have published the efficacy of non-replicating viruses as viable immunotherapies in different cancer models.
