Aromaticity in Fully π-Conjugated Open-Cage Molecules.

J Am Chem Soc

December 2022

Department of Chemistry, National University of Singapore, 3 Science Drive 3, Singapore 117543, Singapore.

Although aromaticity in 2D π-conjugated monocyclic and polycyclic molecules has been intensively studied, aromaticity in 3D fully π-conjugated molecular cages remains largely unexplored mainly due to the synthetic challenges. Herein, we report the facile synthesis of a π-conjugated molecular cage () containing four dimethylmethylene-bridged triphenylamine (DTPA) units via platinum-mediated assembly of four molecules of a pinacol borate trisubstituted DTPA derivative, followed by reductive elimination. has an open-cage structure, consisting of two isomeric trimers in - and -configurations, and an additional macrocycle across four DTPA units.

Platinum chemotherapies are highly effective cytotoxic agents but often induce resistance when used as monotherapies. Combinatorial strategies limit this risk and provide effective treatment options for many cancers. Here, we repurpose atovaquone (ATQ), a well-tolerated & FDA-approved anti-malarial agent by demonstrating that it potentiates cancer cell death of a subset of platinums.

Platinum is a precious and versatile metal and has played important roles in various fields of chemistry ranging from functional materials to anticancer drugs and catalysis. This account highlights an adventurous journey of platinum from promoting phosphorescence to catalyzing a C-H functionalization reaction, or you may consider it our research adventure from phosphorescent materials to catalysis. Interestingly, this journey is driven by a series of "bad" reactions, and of course more importantly by our curiosity about the "bad" reactions.

The reactions of [PtCl2(NCR)2] with telluroethers (ArAr'Te) in organic solvents have been investigated. The reactions in dichloromethane yield [PtCl2(TeArAr')2], while those in tetrahydrofuran (THF) give different products depending on the steric demands of the aryl groups on tellurium, the molarity of the reactants, and the reaction conditions. The reactions between [PtCl2(PhCN)2] and TeArAr' in 1:1 molar ratio at room temperature in THF yield several products, like [PtCl2(TeArAr')2] (Ar/Ar' = Ph/Ph, o-tol/Mes, Mes/Mes), [PtCl2(PhCN){NC(O)Ph[TeMes(o-tol)]}], and [PtCl2{NC(O)Ph(TeMes2)}2].

The synthesis of platinum-acridine hybrid agents containing carboxylic acid ester groups is described. The most active derivatives and the unmodified parent compounds showed up to 6-fold higher activity in ovarian cancer (OVCAR-3) and breast cancer (MCF-7, MDA-MB-231) cell lines than cisplatin. Inhibition of cell proliferation at nanomolar concentrations was observed in pancreatic (PANC-1) and nonsmall cell lung cancer cells (NSCLC, NCI-H460) of 80- and 150-fold, respectively.
