We describe a general method for the mimicry of one face of an alpha-helix based on a terphenyl scaffold that spatially projects functionality in a manner similar to that of two turns of an alpha-helix. The synthetic scaffold reduces the flexibility and molecular weight of the mimicked protein secondary structure. We have applied this design to the development of antagonists of the alpha-helix binding protein Bcl-x(L). Using a sequential synthetic strategy, we have prepared a library of terphenyl derivatives to mimic the helical region of the Bak BH3 domain that binds Bcl-x(L). Fluorescence polarization assays were carried out to evaluate the ability of terphenyl derivatives to displace the Bcl-x(L)-bound Bak peptide. Terphenyl 14 exhibited good in vitro affinity with a K(i) value of 0.114 muM. These terphenyl derivatives were more selective at disrupting the Bcl-x(L)/Bak over the HDM2/p53 interaction, which involves binding of the N-terminal alpha-helix of p53 to HDM2. Structural studies using NMR spectroscopy and computer-aided docking simulations suggested that the helix binding area on the surface of Bcl-x(L) is the target for the synthetic ligands. Treatment of human embryonic kidney 293 (HEK293) cells with terphenyl derivatives resulted in the disruption of the binding of Bcl-x(L) to Bax in intact cells.

The reaction of terrylene in p-terphenyl with molecular oxygen is reinvestigated by TIRF-microscopy with λexc = 488 nm or λexc = 561 nm and 488 nm. A similar range of fluorescent products is obtained under both experimental conditions with a reaction quantum yield Φr > 10-7 for those molecules which undergo the photoreaction. The majority of these oxygen-susceptible molecules reacts via an electronically relaxed, dark intermediate, presumably an endoperoxide, with a lifetime of ~ 20 s.

Intramolecular aromatic oxidative coupling of 3,6-bis(m-terphenyl-2'-yl)carbazole provided a bis(m-terphenyl)-fused carbazole, while that of 3,6-bis(m-terphenyl-2'-yl)-1,8-diphenylcarbazole afforded a bis(quaterphenyl)-fused carbazole. Borylation of the latter furnished a B,N-embedded helical nanographene binding a fluoride anion via a structural change from the three-coordinate boron to the four-coordinate boron. The anionic charge derived from the fluoride anion is stabilized over the expanded p-framework, which leads to the high binding constant (Ka) of 1 × 105 M-1.

In this study, we report the first synthesis of nocarterphenyl A (), characterized by a thiazole-fused -terphenyl framework. Furthermore, we synthesized 18 additional analogs, including the naturally occurring compound 5-methoxy-4,7-bis(4-methoxyphenyl)benzo[]thiazol-6-ol (), employing a similar synthetic approach.

Reactions of a m-terphenylhydridostannylene with β-diketiminato magnesium and calcium hydrides provide bis-μ-hydrido species, the heterobimetallic constitutions of which are maintained after the addition of THF donor solvent. In both cases, reactions with hex-1-ene result in the formation of tetravalent organostannyl alkaline earth derivatives. Whereas the magnesium reagent undergoes facile twofold addition, the calcium-centered process is arrested after a single alkene reduction event.
