Aim: To study the role of adaptor protein Bam32 in B cell antigen receptor (BCR) signaling cascades.

Methods: Using full length Bam32 as bait, yeast two-hybrid technique was used to screen the protein that could interact with Bam32. The interaction was further confirmed by co-transfection of 293T cells and coimmunoprecipitation.

Results: Protein tyrosine kinase Lyn was one of the strong positive clones identified by the yeast two-hybrid screening. This interaction was further confirmed in 293T cells by co-transfection and coimmunoprecipitation. By using specific anti-phosphotyrosine antibody, it was found that Bam32 could be phosphorylated by Lyn.

Conclusion: The interaction of Bam32 with Lyn leads to Bam32 phosphorylation, which might play an important role in activating downstream signaling molecules.

Insulin resistance was considered to be the most important clinical phenotype of type 2 diabetes (T2DM). Almond is a widely-consumed nut and long-term intake was beneficial to alleviating insulin resistance in patients with T2DM. Hence, screening of anti-diabetic peptides from almond proteins was feasible based on the effectiveness of peptides in the treatment of T2DM.

Palmitate potentiates the SMAD3-PAI-1 pathway by reducing nuclear GDF15 levels.

Nuclear growth differentiation factor 15 (GDF15) reduces the binding of the mothers' against decapentaplegic homolog (SMAD) complex to its DNA-binding elements. However, the stimuli that control this process are unknown. Here, we examined whether saturated fatty acids (FA), particularly palmitate, regulate nuclear GDF15 levels and the activation of the SMAD3 pathway in human skeletal myotubes and mouse skeletal muscle, where most insulin-stimulated glucose use occurs in the whole organism.

Background: Resistance to temozolomide (TMZ) remains is an important cause of treatment failure in patients with glioblastoma multiforme (GBM). ADAR1, as a member of the ADAR family, plays an important role in cancer progression and chemotherapy resistance. However, the mechanism by which ADAR1 regulates GBM progression and TMZ resistance is still unclear.

Purpose: Cardiomyocyte death is a major cytopathologic response in acute myocardial infarction (AMI) and involves complex inflammatory interactions. Although existing reports indicating that mixed lineage kinase domain-like protein (MLKL) is involved in macrophage necroptosis and inflammasome activation, the downstream mechanism of MLKL in necroptosis remain poorly characterized in AMI.

Methods: MLKL knockout mice (MLKL), RIPK3 knockout mice (RIPK3), and macrophage-specific MLKL conditional knockout mice (MLKL) were established.
