Classical swine fever (CSF) is a highly contagious disease caused by classical swine fever virus (CSFV). NS5A, a non-structural protein of CSFV, plays an important role in regulating viral replication and protein translation. The purpose of this study was to investigate the effects of the CSFV NS5A protein on key proteins in the mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K)-mechanistic target of rapamycin (mTOR) pathways in porcine macrophages.

The classic subgroup A (ALV-A) and newly emerging subgroup K (ALV-K) of avian leukosis virus are two major pathogens responsible for avian leukemia in chickens, posing substantial threats to global poultry industry. Both viruses share a Tva protein encoded by the tva gene as a receptor to gain the entry into the host cells. In this study, we described the identifications of two alleles of the tva receptor gene in Qingyuan partridge chicken, which possesses an 11-nucleotide (GCTGCCCACCC) insertion and a 6-nucleotide (ACCTCC) deletion independently located in exon 1 of the tva receptor gene.

Piggybacking on nature: exploring the multifaceted world of porcine β-defensins.

Porcine β-defensins (pBDs) are cationic peptides that are classically associated with the innate immune system. These molecules yield both antimicrobial and immunomodulatory properties, as evidenced by various in vitro and animal trials. Researchers have revealed that enhancing pBD expression can be achieved through dietary components and gene editing techniques in pigs and porcine cell models.

A classical swine fever (CSF) vaccine combining high potency and an immunological marker for differentiating infected animals from vaccinated animals facilitates disease control and provides proof of eradication to promote international pig trade. Previously, CSF virus (CSFV) recombinant live vaccine strains, guinea-pig exaltation of Newcastle disease virus-negative strain vaccine [vGPE (genotype 1.1)], were developed with the E gene replaced by non-CSF pestiviruses (Pronghorn antelope or Phocoena pestiviruses).

For a better understanding of classical swine fever (CSF) pathogenesis, a transcriptomic analysis was performed using porcine bone marrow (BM)-derived antigen-presenting cells (APCs) infected ex vivo with two different cDNA-derived classical swine fever virus (CSFV) strains, the low-virulence Pinar de Rio (vPdR-36U) or the lethal vPdR-HK-5U. The transcriptomic profile of vPdR-36U- or vPdR-HK-5U-infected versus noninfected cells revealed 946 and 2643 differentially expressed genes (DEGs), respectively. The upregulation of ISG15, CXCL-10, ADAM8, and CSF1 was found after infection with vPdR-36U, which could contribute to the generation of mild CSF forms.
