Thermus thermophilus HB27 laccase (Tth-Lac) is a thermostable enzyme that contains a β-hairpin (Ala292-Gln307) covering the substrate entrance. We analyzed the role of this β-hairpin in the enzymatic activity of Tth-Lac through three β-hairpin mutants: two variants without the β-hairpin (C1Tth-Lac and C2Tth-Lac) and one with a partially modified β-hairpin (P1Tth-Lac). Enzymatic activity was assayed with different substrates with and without copper.

Probiotics and their postbiotics have the potential to improve the health and growth performance of piglets, which has brought them widespread attention in the post-antibiotic era. In the present study, the effects of dietary supplementation of postbiotics on the growth performance, intestinal flora structure and plasma metabolome of weaned piglets were investigated. A total of 816 healthy male piglets with uniform weight were divided into two treatment groups: piglets in the control (CTR) group were fed with a basic diet, and the ones in the LAC group were fed with the basic diet supplemented with 500 mg/kg postbiotics.

This review starts with a historical overview and a description of the classical example of allostery – hemoglobin – and other well-known examples (aspartate transcarbamoylase, Lac repressor, kinases, G-protein-coupled receptors, adenosine triphosphate synthase, and chaperonin). We then discuss fringe examples of allostery, including intrinsically disordered proteins and inter-enzyme allostery, and the influence of dynamics, entropy, and conformational ensembles and landscapes on allosteric mechanisms, to capture the essence of the field.
